Objective:To explore the expression, distribution, and dynamic change of MMP10 in endometria of early pregnant mouse, with the purpose of providing experimental and theoretical support for the exploration of blastocyst implantation.Methods:1. Endometrium from day 3-7 in early pregnancy mice were used to following study. Both non-pregnant (d0) and pregnant mice (d3, d4, d5, d6,d7) were randomly divided into 6 groups.2. The level of the expression of MMP10mRNA was detected by RT-PCR and in-situ hybridization.3. The level of the expression of MMP10 was tested by Western blotting and the Immunohistochemistry.Results:1. MMP10 is expressed in the endometrium of both non-pregnant and early pregnant mice, with the expression level of the latter higher than that of the former(P<0.05). 2. The size of the amplified fragment of RT-PCR is the same as the expected value (210bp),the expression of MMP10mRNA in early pregnant group increased significantly as compared with that in unpregnant group, also, the expression strengthens gradually from early implantation to late implantation, with day 6 and day 7 having the strongest expressions which are significantly different from those of other days(P<0.05).3. The results of in-situ hybridization are basically consistent with those of RT-PCR.4. IHC shows that the expression of MMP10 in early pregnant group increased significantly as compared with that in non-pregnant group, the expression of MMP10 strengthens gradually from early implantation to late implantation, with day 6 and day 7 having the strongest expression which is significantly different from those of other days(P<0.05). Brown granules are found to exist in cytoplasmic of stromal cells, Luminal epithelium, and glandular epithelium, but mainly exist in stromal cells, with Cytosolic having a larger concentration and nucleus less.5. The results of Western blotting is basically consistent with those of IHC.Conclusion:The dynamic expression and distribution change of MMP10 in the endometrial stroma cell of early pregnant mice suggest that the high expression might facilitate the degradation of extracellular matrix, the invading of embryonic trophoblast cells, and the participation in the decidualization of endometrial during the implantation and the early proliferation and differentiation of embryos.
