Follicle stimulating hormone (FSH) is a glycoprotein hormone secreted by the anterior pituitary and composed of glycosylatedα-subunit andβ-subunit which are non-covalently bounded. FSH plays an important part in animal reproduction and the preparation of FSH have been widely applied to improve the reproductive capacity of livestock .At present most of FSH are distilled from livestock's pituitary which are mixed with Luteinizing Hormone (LH) and easily infected by pathogeny .A lot of FSH genes have been cloned by many researchers and expressed out of body in mammalian cell system,but the level was much low .The bioreactor of mammalian mammary gland was accepted to increase the level of expression of interesting genes.On the basis of former, this research have constructed the transient expression vector of FSH and Its long-acting analogous genes in mammary gland and depending on the transient expression in mammary gland to exmine the rationality and validity of the construction of vector ,then lay the foundation for researching transgenic animal and improving the application and development of transgenic technology.Designing specific primers containing corresponding restriction enzyme sites, FSHβ-CTP,FSHβ-CTP-αcontaining special restriction sites were amplied by two PCR respectively ,with the template of pVITROFSHβ-CTP-α.
