| Insulin-like growth factor-I (IGF-I) is a kind of multipotential regulatory factor that has wide biological effects. The factor can promote proliferation and differentiation of many kind of cell. As a result it has been put in use to treat many diseases such as diabetes, osteoporosis, disease of neuromus-cular and so on .Unfortunately the quantity of IGF-I in blood is very limited, moreover it mainly exists in the form of inactive complex. Consequently, it is difficult to get IGF-I in large scale for the scientific research and clinic uses. Therefore it has become the focus of attention to produce enough IGF-I for use by using genetic engineering technique.The result of experiment shows that the engineered bacteria BL21(DE3)/ pExSecI/IGF-I were induced to express IGF-I with high rate by IPTG (0.8 mmol/L ) at 37℃ for 3 hours. The recombinant protein was over expressed as inclusion bodies. After the inclusion bodies have been solubilized by denaturing agent (8 mmol/L Urea) , it was renatured by step wise dialysis, then purified through HiPrep 16/60 Sephcryl S100 High Resolution, rIGF-I was purified to 90% of purity. Western Blotting test proved that the recombinant protein showed immuno-activity, and MTT assay showed that rIGF-I can promote proliferation of NIH3T3 cell.Mechano Growth Factor (MGF) is derived from the insulin-like growth factor (IGF-I). MGF is expressed by mechanically overloaded muscle and is involved in tissue repair and adaptation. These donate nuclei to the muscle fibers that are required for repair and for the hypertrophy processes which may have similar regulatory mechanisms. Muscles in the elderly are unable to upregulate MGF in response to exercise.
