This laboratory cloned the porcine growth hormone (pGH) gene, and it was correctly inserted into a eukaryotic expression vector VR1020 and got the expression plasmid VpGH. We cloned the porcine interlukin-6 (pIL-6) gene, and got the expression plasmid VpIL-6 in the same method. The recombinant plasmid was transformed into TOP10F', and it was identified by plasmid PCR and enzyme digestion. Cation-Liposome was prepared by the method of the thin film, wrapping VpGH genes, VpIL-6 plus VpGH genes with this kind of liposome. Using the four disposals, we injected Kunming mouse of 21days old. The mouse was weighted after injection 0, 7, 14, 21, 28, 35 days. At the same time, each week we drew blood by the method of cutting their tails and separated blood serum to analyze the content of GH and the level of IgG with ELISA, and examined the quantity of the white blood cell, red blood cell, lymphocyte, mononuclear cell, neutrophil. The result revealed that indices of the rats disposed by co-expression of VpIL-6 and VPGH genes are evidently boosted up compared with the ones of rats disposed by VpGH genes solely (P<0.05). While the response of cell immunity and body fluid immunity does not show remarkable difference.From the result of the experiments on the mouse, we used the same experiment on the pig.
