We chose Selaginella tamariscina (Beauv.) Spring which is a kind of resurrection plants as our experiment sample. With 5%, 10%, 15% PEG to imitate drought stress, we researched the physiological change of S. tamariscina (Beauv.) Spring with the theory of plant Physiological ecology. And then we made use of the functional proteomics to explore the varieties protein of S. tamariscina (Beauv.) Spring which was under the PEG stress. The purpose of our experiment was that discussing the relation of drought-resistance and plant protein under PEG stress, and atempted to reveal mechanism of drought-resistance of S.tamariscina (Beauv.)Spring in molecular level. At the same time, we controled and optimized the experimental conditions of the two-dimensional gel electrophoresis system.The research determined the content variety of proline in leaves of S. tamariscina (Beauv.) Spring with deal time under different PEG concentration stresses, as well as the permeability of plasma membrane, SOD activity and POD activity were measured. By combination and optimization of various conditions, a sample preparation method from leaves of S.tamariscina (Beauv.) Spring was established. And then we founded a set of method of two-dimensional gel electrophoresis, the discrepancy spots which were in the comparison and drought stress, were analyzed by silver staining. Whereafter we accumulated the discrepancy spots by the coomassie brilliant blue staining, and then 18 spots were obtained, mapping peptide mass fingerprinting of protein spots by MALDI-TOF-MS, 11 spots were identified with new protein, and 7 spots were identified with known proteins from protein database using Mascot, physiological functions of 7 proteins were analysed at the course of S.tamariscina (Beauv.) Spring drought stress.
