The phylogenetic relationships of Calycanthaceae were investigated by means of molecular systematics', which was based on three DNA sequences from three genomes, including chloroplast DNA trnL-F, nuclear 18S ribosomal DNA, and mitochondrial matR gene. The present study includes reconstruction the phylogeny of Calycanthaceae, discussion the systematic position at family level, and relationships among genera. In addition, a cladistic based on the morphology of Calycanthaceae was carried on. The results were summarized as follows: 1.18S rDNA sequence analysisThe strict consensus trees of 18S rDNA made by BI and MP are similar, with only a few differences in bootstrap value or Bayesian posterior probability for some clades. The topological differences observed between the MP and BI analyses focused on the internal clades of Ranales (Ranunculaceae) and Rosales (Rosaceae). With the method of BI, they are sister group (BS=68%) while MP doesn't support their relationship. However, the monophyly of Calycanthaceae was supported in the strict consensus tree of both analyses.Ch.campanulatus var.guizhouensis, Ch.nitens and Ch.parecox are sister groups with the bootstrap values of 98% (MP) and 82% (BI), Ch.nitens and Ch.parecox are sister groups. The closest relationship of Calycanthaceae and Idiospermaceae are supported by 99% and 77% respectively.2. Mitochondrial matR gene sequence analysisIn the strict consensus tree of matR, the topological difference observed is little. Magno-liales (Magnoliaceae), Rosales and Ranales are on the base of the tree, their relationships are unclear in MP, but in BI Ranales and Rosales are closest and they are the sister groups of Magnoliales, while all the three orders is the sister group of Laurales.But they both support the clade of Calycanthaceae and Idiospermaceae (BS=100%).The monophyly of Calycanthaceae is supported by 86% and 97%.3. cpDNA sequences analysisIn the strict consensus tree of trnL-F, the topology of the tree are similiar except for a few differences in bootstrap support or Bayesian posterior probability for some clades.The Calycanthaceae are divided into two groups,one including Sinocalycanthus,C.floridus and...
