Selenium is a nessassary trace element for human nutrition, Soybean oligo-peptide has many biological functions. If We can efficiently band the study on selenium and soybean oligo-peptide together, explor Se-contained soybean oligo-peptide, it may be more valuable.We choosed the Se-contained soybean,which came from Enshi in Hubei—the capital of selenium in the world, Se-contained soybean protein has been extracted from it, than used protease We choosed due to experiments to enzymolysis the Se-contained soybean protein. Proccessed separation and purification, We gained Se-contained soybean oligo-peptide, its molecular weight is below 1000.The main results of this study are as follows:According to references and their capacities to enzymolysis the Se-contained soybean protein and their costs, We choosed Bacillus subtilis protease and papain as the tool enzyme in this study.We investigated the optimum condition of using the papain and Bacillus subtilis protease to enzymolysis the Se-contained soybean protein, concentration of enzyme, temperature, pH and concentration of substrate affect the reaction of enzymolysis. Due to the degree of their affection to the reaction of enzymolsis, those factors take turus: concentration of enzyme > temperature > pH >concentration of substrate. the optimum condition of using the papain to enzymolysis the Se-contained soybean protein is: concentration of enzyme 15000u/g, temperature 55℃,pH7.0, concentration of substrate 2%, need 3h; the optimum condition of using the Bacillus subtilis protease to enzymolysis the Se-contained soybean protein is: concentration of enzyme 8000u/g, temperature 55℃,pH7.0, concentration of substrate 2%, need 4h.We investigated the optimum condition of using the mixture of papain and Bacillus subtilis protease to enzymolysis Se-contained soybean protein .The appended sepuence of proteases nearly have nothing affection to the DH, the optimum condition of mix-enzymolysis is: temperature 55℃, pH7.0, concentration of substrate 2%.
