A more effective and more widely used method is established to display the microtubule organelle of ciliates with FLUTAX, based on the procedure described by Arregui et al. Four main added steps are coating slides with polylysine, incubating cells with saponin, fixing cells with polyformaldehyde and infiltrating cells with Triton X-100.According to the improved method of FLUTAX, using flutax as the probe, tracked and marked the microtubule organelles and the structure microtubles as well as their changes at different paste of life cycle of a kind of ciliates hypotrichida stein (Euplotes sp.) .In the progress of agamogenesis, ventral ciliature, including adoral zone of membranelles, paraoral membrane, frontal cirri, ventral cirri, transverse cirri and caudal cirri disorganized, new ciliature grow on definite position of cell cortices. From the creation, disposition and movement of the corresponding ciliature anlagens, to finally getting the former and latter new ciliature, the formation of the new construction is going together with the disorganization of the old one. During the growing of dorsal ciliatures, the new ciliate anlagens grow at the back of the old one, and finally becoming tow new set of dorsal kinety, the former one and the latter one. According to the differentiation feature of the microtubule organelles in different physiological states, the writer assumes that microtubule organelle made a continuous change in different paste of ciliate's life cycle, and so it's the foremost important structure of cell. In the progress of shape creation and structure disposition, former microtubule structure may have physical contribution to the building of new construction, and it has the function of structure inducement and pattern location.This research also used a tubulin antibody and y tubulin antibody to mark growing progress of microtubule organelles in euplotes in different period, a tubulin antibody has strong specificity. By combining with a tubulin in the organelles and its skeleton, it tracked and marked the creation, movement and placement of ciliature skeleton and organelle microtubles during the cell cycle.
