Comparing with the multicellular life-forms, Hypotrichous ciliates have complex microtubule structures, the study of microtubule-like organelle of ciliates has been one of the important research fields in protozoology and cell biology , several methods have been used to display these microtubule-like structures. But for a long time, there were not good methods to display microtubule-like structures technically. Traditionally, sliver impregnation was used to display ciliates' kinetosomes, but the structure were unclear; displaying cilia and adjunctive structure by protargol technique while the inner microtubule can not been displayed. To further investigate the characters and components of cytoskeleton of ciliates, using the method-FLUTAX and immunifluorescence technique to observe microtubule-like organelle and microtubulin of Pseudokewnopisis rubra and Tachysoma in different stages of the life cycle ; by means of western blotting, to investigate the α , β,γ- microtubulin of two kinds of ciliates. The results are showed as fellows:1. microtubule-like skeleton of Pseudokewnopisis rubra and Tachysoma in different physiological statesWith the method-FLUTAX, the shape and differenation of the microtubule and microtubule-like structure of Pseudokewnopisis rubra and Tachysoma in different physiological states, can be visioned clearly. During the binary fission of the cell, the new cirri kinetosomes come into being in certain given position of the ventral side, they develop into cirri and migrate and divide into two sets of cirris, at last take the place of the old ones .on the ventral side of cells, the adoral zone of membrane(AZM), the underlating membrane(UM), froto-ventral and transverse cirri(FVC,TC) and caudal cirri migrate and then are disintegrated .
