The specific expression of foreign gene depends on the proper adjustment of promoter, especially some anther-tissue-specific promoters which are the key factor to create crop male sterility line and corresponding restore line. In research the plant whose anther specific promoter was tagged was successfully chosen out from the transgenic tritordeum by non-promoter uidA tactic, and the inheritance stability of foreign locus was researched, which made a good preparation for the subsequent isolation of promoter. The results are as followed: 1) Different tissues of the transgenic plants in different periods were collected to analyse the patterns of uidA expression by histochemistry Gus assay. RT-PCR was applied to make ulteriorly sure of the characteristic of the promoter from the level of DNA transcribing. It was verified that the patterns of uidA expression of the selected plant was the same as that of the T0 generation , only confined in the anther primordial and pollen; 2) The locus was inherited at a low frequency, namely, a majority of foreign Gus copies were absolutely lost, not in accordance with the rule of dominant character: recessive character (3:1), but the patterns of Gus expression was highly stable, which revealed that the structure and function of its inherent promoter is intact and precise respectively, although the transgenic crop has gone through several generations; 3) The foreign Gus was inserted behind the crop interior promoter, which affected the function of the gene related to the gametal development or pollination process, which accordingly resulted in the abnormal development of the gametal. Even if the gametal could temporally complete normal development, the subsequent pollination processes, including the cell division of zygote cell, the differentiation of special cell or the formation of tissue and organ, and so on, were affected, which may make it impossible to get the transgenic homozygote.
