Tartar buckwheat with high pharmaceutical and edible values is a dicotyledonous plant and specially cultivated in China, which is adapted to cool climates and short-growth period. The bioactivity components in tartar buckwheat with antihypertension, antihyperlipemia and antihyperglycemia mainly is Flavonoids, the most important one of which is rutin. In this thesis effects of different extraction conditions on the content of rutin in Fagopyrum tataricum(L) Gaertn were studied, and some rutinos ramifications were synthesizede by using rutin-degrading enzymes(RDEs). In addition, the progress of study on this plant was summarized . Firstly, Effects of different extraction conditions on the content of rutin in Fagopyrum tataricum(L)Gaertn were studied by HPLC,including time of touched in water, ethanol concentration, temperature and pH. The results showed: That tartar buckwheat had high active rutin-degrading enzymes; That the content of rutin was remarkably affected by the time of water touching. That the content of rutin decreased from 79.72% to 32.21% during touched in water one minute, i.e. about 60% rutin had been hydrolyzed, after fifteen minute, the content of rutin changed very slowness and was about 14.37%; Ethanol of different concentrations all have inhibiting effect on rutin-degrading enzymes, the content of rutin increase with the raise of ethanol concentration. That the activity of rutin-degrading enzymes was evidently restrained by more than 40%(V/V) ethanol, the most evident one of which was 90%(V/V). Inhibiting effect of temperature on rutin-degrading enzymes was unconspicuous The activity of the rutin-degrading enzymes was restrained just at T≥60℃, and the content of rutin began to increase slowly at the same time, increase rapidly at T≥70℃and was up to 80% of original value at T=90℃. The rutin-degrading enzymes had well acid-resistance and alkali-resistance in the range of pH1.0 to pH11.0. In this range ,the content of rutine changed unconspicuously and the average was about 12%. Secondly, the change of the content of rutin during the germination of buckwheat seeds was studied. The result of HPLC analysis showed that the rutin-degrading enzymes was restrained in growth period of buckwheat seed; the content of rutin increased with augment of day and length of seeding. that the content of rudin increased by two fold in buckwheat seeding after ten day (the length was 9 cm). Finally, synthesizeing ?-hydroxy rutinoside using rutin-degrading enzymes was exploringly studied. Separate and purify Rutin-degrading enzymes from buckwheat. Using rutin and rutin-degrading enzyme as substrates got five compounds, and analyzed configuration of those reactants by using IR, FAB-MS, 1HNMR, 13CNMR and DEPT. Conclude they were rutinos, methyl-?-rutinoside, ethyl-?-rutinoside,propyl-?-rutinoside and isopropyl-?-Rutinoside. These results were important to the extraction of rutin from Fagopyrum tataricum(L) Gaertn and its food production, and offer one way to prepare of rutinos ramification..
