With the development of science and technology, science of biology has become more and more important. As a main carrier of live information-DNA, it has excited great interest across of wide range of scientific enthusiasm among scientists in recent years. This work studies the interaction between Eu3+ - rutin complex and DNA using electrochemical and fluorimetric methods. The main content consists of four parts.Part I . Development in the interaction of metal complexes with DNAThis review provides a simple introduction to the development of the interaction between metal complexes and DNA, and provides some basic theory related. Reports about great enthusiasm in the synthesis of the complexes of rare earth medicine, the interaction of the rare earth complexes with DNA are very few in the world. It is expected to explore the mechanisms of interaction that are used to offer an opportunity to understand which parameters affect the binding mode and binding affinity to DNA.Part II. The electrochemistry investigation of rutin and application inanalysisAn electrochemical investigation for rutin on the glassy carbon electrode is presented. A adsorptive process is observed. The maximum coverage on the electrode surface is 5.09 × 10-10mol/cm2. corresponding to a benzene ring with 3'. 4'-dihydroxides adsorbed on the electrode surface, the other parts of rutin stay away from the surface. In the meantime, a new electrochemical method for determining rutin is presented. In a medium of 0.1M acetate buffer solution(pH=4.46). a very sensitive polargraphic adsorptive wave is observed by using a differential pulse voltammetry at about 0.29V(vs. Ag/AgCl). The linear relationship between the peak current and the concentration of rutin exists in a range from 3.28× 10-7M to 3.28× 10-5M. The detection limit of rutin is down to 2.51 × 10-8M. This method has been applied to determine rutin in several samples of Chinese medicines. The results are satisfactory.Part HI. The electrochemistry investigation of the binding mode of Eu3+-rutin complex to DNAThe interaction of the Eu3+-rutin complex by DNA is studied. Studies involved the interaction of the Eu3+and rutin, Eu3+-rutin complex and DNA. The electrochemical investigation for Eu3+-rutin complex on the glassy carbon electrode is presented, revealing that a intercalation interaction between double stranded DNA and Eu3+-rutin complex under the condition of little acid. The intrinsic binding constant is 1.42×104 M-1, and binding site size is 2.Part IV. The fluorescence investigation of the interaction between theEu3+-rutin complex and DNAThe interaction of the Eu3+-rutin complex with calf thymus DNA was studied using fluorescent method. The mode of Eu3+-rutin complex binding to DNA by intercalation was observed. A more direct observation the intercalation interaction by addition the Eu3+-rutin complex into DNA under the optimal conditions(0.1M acetate buffers. PH=5.69) using fluorimetric method was presented. According to the spectral excitation and emission spectrum of DNA and guanine. the complex intercalate into the G-C base pairs of DNA.
