A great deal of factors has multiple effects on the success rate of cell nuclear transfer. Besides the effect of maintaining ploidy of reconstructed ovum core and reprogramming the donor nuclei, treatment method of oocyte , micro- operating technology, different cytoplast recipient and different donor nuclei and its state have important influence on construct development of embryo. The recipient is oocytes in metaphase- II, donor nuclei is the granule cell in this experiment. Influence factors on reconstructed embryo in nuclear transfer technology of mice were reserched, The results as follows.1, The cumulus-enclosed oocyte complexes treated with hyaluronidase , the concentration and treated time both influence the development of reconstruct embryo. With move ovum tube blowing and beating for 2min repeatedly , can obtain better reconstruct embryo development rate (80.8%).2, This experiment compare different parameter of electricity activate and age influence on the oocyte activate.Oocytes treated with same pulse strength , pulse duration, pulse times, The oocytes at 17h can get high activating rate (71.7%) and development rate (84.8% /39.4%).In the same age, pulse duration and pulse times, The oocyte activate rate reached tiptop(72.7%) when the pulse strength at the 100 V/mm, 2-cell and blastula develope rate also reached peak value (85.0% /35.0%), so this pulse strength was suitable for oocytes activation.In the same age, pulse strength and pulse times. Pulse duration at the 80μs, the oocyte activate rate reached tiptop(76.7%), 2-cell, blastula develope rate both reach peak value (80.4% /34.8%), so this pulse duration was suitable for oocytes activation..In the same age, pulse strength and pulse duartion, the activate rate of 3 times' pulse is higher than 2 times' pulse, while it was just reverse of 2-cell and blastula develope rate (70.3% /29.7%). So 2 times' pulse was suitable for oocytes activation.
