Based on morphological taxonomy , we applied a new maker of ISSR and conventional one of RAPD to study the molecular taxonomy of 22 isolates of the genus Pleurotus and one of Agaricus bisporus . The results of this study are as follows:The result of morphological characters of mycelium and sporocarp showed that Agaricus bisporus and genus Pleurotus is definitely different, P. abalonus is differ from the others isolates of genus Pleurotus and alone constitutes a species, and NO. 16 P. sp. and P. sajor-caju are so similar that they can made up of one species .Other isolates are indefinite with P. ostreatus and P. floridanus.The 478 bands were obtained which were amplified by the genome of isolates used in study and seven primers which were screeened from RAPD primers of the S series. Cluster analysis showed that at 71% similarity level, all tested isolates could be clusted into eight groups, i.e. I NO.1-3 P. ostreatus , NO.13-14 P. floridanus, NO.18 P. sp. and P. corticatuus. â…¡ P. sapidus, NO.15P. sp and NO.17 P. sp. â…¢ NO.11 P. sp. â…£ P. ferulae, P. nebrodensis and NO.6 P. ferulae. â…¤ P. cornu- copiae, P. limpidus and NO.9 P. sp. â…¥ P. sajor-caju and NO.16 P. sp. â…¦ NO.20 P. sp and NO.22 P. sp . â…§ P. abalonu.The 119 bands obtained from the result of ISSR-PCR . Cluster analysis based on similar coefficient showed that at 71% similarity level, all tested isolates of genus Pleurotus can be divided into four groups. i.e. â…¡ P. ferulae , P. nebro- densis and NO.6 P. ferulae. â…¢ P. sajor-caju and NO.16 P. sp. â…£ P. abalonu . the left constituted group I . At last the 1.3 Kb ISSR fragment of NO.17 isolate was obtained from 1.5% agarose gel ,cloned and sequenced, in which the (TATG)_n motif was found .In conclusion, the taxon of genus Pleurotus have been investigated systematically by using morphological character, makers of RAPD and ISSR. The results suggested that P. abalone alone constitutes a group for the reason of having further genetic relationship with other isolates, NO. 1-3 P. ostreatus and NO.13-14 P. floridanus had very intimate genetic relationship and constituted one species with different names, and P. sajor-cajuand NO. 16 P. sp. had terribly closer relationship and can be composed the species P. sajor-caju, as well as P. ferulae, P. nebrodensis and NO.6 P. ferulae, the left isolates such as NO.15 P. sp., NO.17-18 P. sp. and P.sapidus also have closer genetic relation-nship and may constitute a species.
