DNA molecular marker is a new advanced technology applied in molecularbiology and biotechnological research. It has some particular advantages comparedwith other markering technology, such as morphologic marker, cytological marker andbiochemistry marker. At present, there are five major methods, RFLP, RAPD, AFLP,SSR and ISSR used to detect the DNA variance. Currently, the technology of DNAmolecular markers are being widely applied in the areas of plant research ingermplasm classification and reservation, genetics study, gene mapping and markerassistant breeding.Thellungiella salsuginea (salt cress ),belonging to Thellungiella of Cruciferae, is anannual herb. It is a close relative of Arabidopsis. Studies showed that 300mM NaClwas a salinity threshold for salt cress growth which decreased the survival rate by50% relative to the medium without salt, 100~150mM NaCl was the optimum growthsalinity for salt cress. As we know, a model plant would be needed that had desirabletraits, i.e. small size, short life cycle, self-pollination, and high seed number, andfavorable genetic traits such as self-fertilization, a small genome, efficienttransformation, and mutagenesis. Thellungiella salsuginea is one of the halophyteswho meets all of these criteria.Few reports concern research about Thellungiella salsuginea. In the paper, wehave collected seven populations of Thellungiella salsuginea from China and onepopulation from North America, and we have studied the genetic diversity of thepopulations by means of SSR and RAPD, which can prepare for the genetic mapping.The genetic diversity of Thellungiella salsugine populations was investigated byRAPD. 23 primers were chosen for amplification among populations because theyshowed strong, reproducible amplification and distinct polymorphisms from over 400arbitrary primers.
