Four kinds of main Amanita peptide toxins were identified by means of RP-HPLC, MS, UV spectrograph. The method of RP-HPLC determination of alpha-amanitin in the body fluid had been developed. Meanwhile the toxicity of three kinds of main peptide toxins had been analysed. The results are as followings:1. Four kinds of main peptide toxins were purified and preliminary determined by means of RP-HPLC, UV spectrograph and MS from Amanita fuliginea. Three of them are Amanin, phalloidin(PHD)and phalloin(PHN). The molecular weight of the last one is 1145.4636 Da.2. The toxicity of Peptide toxins were as following: Amanin is slow function toxin, the white mouse's LD50 is 565μg . kg-1, the death time of the white mouse is 3-7 days, the later period of poison is able to cause the liver to wither. At the same time it can remarkably suppress the growth of the mung bean seed radicle; PHD, PHN are quick-action toxins, the white mouse's LD50 is 2000μg . kg-1 and 1500μg . kg-1 respectively, the death time of the white mouse is 2-4 hours, all can cause the liver turned escent hyperemia, PHD can suppress the elongation of the mung bean seed radicle.3. Amanin and PHD were screened in biological activity with the ELISA method, which showed no inhibitory action to PP1, PAI-1, CDC25, Cat-K and Cat-B.4. The RP-HPLC method for the rapid determination of α -amanitin in the body fluid of patients caused by poisonous mushroom was developed.
