To investigate the relationships between cell proliferation, apoptosis and the expression of HSP 70, and look for the possible signal pathway, HeLa cells were treated under different factors. Cell proliferation, apoptosis and the expression of HSP70 were determined by MTT assay, gel electrophoresis and Western dot, respectively. This thesis includes two sections.In the first section, the proliferation, apoptosis of HeLa cell and its expression of HSP70 were examined under different heat conditions. The results showed that heat stress caused cell apoptosis. Compared with the control group, treatment with 45 centigrade for 15 min could obviously inhibit the proliferation of HeLa cells and increase the expression of HSP70. With the teperature declined, the affects decreased gradually. The quantity of HSP70 expression was relative to cell's incubation time. In the three treatment group (41, 43 and 45 centigrade), the maximal expression of HSP 70 appeared respectively after 24 h, 24 h and 30 h incubation.In the second section, in vitro experiments were conducted to investigate the effects of Dex, Tween-80, Ca2+, EDTA and Aminophylline on HeLa cell proliferation, apoptosis and its HSP70 expression. The results showed that Dex(10-310-6mol/L) ,Ca2+and H2O2 significantly inhibited the proliferation of HeLa cells in dose-dependent manner; Tween-80 (1%,0.1%) inhibited the proliferation (P<0.01); EDTA (10-2,10-3 mol/L) inhibited the proliferation (.P<0.01); Aminophylline affected the proliferation of HeLa cells in dose-independent manner. Except EDTA, treatment with Dex, Tween-80, Ca2+, H2O2 and Aminophylline caused HeLa cell apoptosis and induced the DNA of cell degradation. As compared with the control group, treatment with H2O2, Aminophylline, Dexamthasone and Tween-80 increased the levels of HSP 70 significantly.
