Objective: To knock down the expression of CyclinE gene in cervical cancer cell by RNAi technology. Study its effect on cervical cancer cell proliferation and cell cycle.Methods: The eukaryotic expression vectors special for knocking down CyclinE gene were constructed and transfected into HeLa cell. The mRNA level of CyclinE gene was detected by reverse transcription polymerase chain reaction(RT-PCR);cell proliferation was analyzed by MTT;change of cell cycle was analyzed by flow cytometry respectively.Results: The eukaryotic expression vector special for knocking CyclinE gene was successfully constructed, it can specially knock down the expression of CyclinE gene. Cell proliferation was inhibited according to the results of MTT. Cells stop at G0-G1 phase and cell number in S phase was sharply decreased after RNAi showed by flow cytometry analysis.Conclusion: The expression of CyclinE gene in HeLa cell can be specially knocked down by RNAi technology and malignant proliferation of cervical cancer cell was inhibited .It suggests CyclinE gene may become a new gene therapy target of cervical cancer.
