ω3 (Omega-3 fatty acid desaturase), CBF3 (C-repeat binding factor) and AtGoLS3 (Arabidopsis thaliana Galactionol synthase 3) are genes, which relate to cold tolerance in higher plants. ω3 fatty acid desaturase gene from Arabidopsis thaliana catalyzes the precursor of 18:2 fatty acid AC to form desarurated fatty acid 18:3. The analysis of the functional domain of this gene shows that its functional domain is full of the histidines, which are capable of binding metal ion. CBF3 is a transcriptional factor, which especially binds to the trans-acting DNA regulatory element of C-repeat/DRE involved in COR gene family. The over-expression of CBF3 gene in response to low temperature induces the COR gene expression. CBF3 gene contains a nuclear localization signal domain (NLS) and an acidic region. AtGoLS3 is a Galactionol synthase, when the plant in the adversity condition of cold stress, the AtGoLS3 is induced to over-expression and causes galactinol and rafifinose increased in vivo.A set of primers, which from the coding frame of the ω3 , CBF3 ,AtGoLS3 genes in Arabidopsis thaliana were designed. ω3, CBF3, AtGoLS3 genes were isolated by the RT-PCR. The isolated sequences are 100% matched the sequences in GenBank. The cDNA length of ω3, CBF3, AtGoLS3 is 1064bp, 861bp, 1012bp, respectively.In the present work, the expression vector of ω3 was constructed named 35S-pGreen II 0029-ω3. This construct uses kanamycin as selection marker. The 35S-pGreen II 0029-ω3 was transformed to Arabidopsis thaliana and the explants (cotyledon and hytocotyl) of Stylosanthes guianensis cv. Reyan No. 5 through Agrobacterium tumefaciens GV3103. As a result, several independent lines of transgenetic Arabidopsis plant and transgnentic calli of Stylosanthes guianensis were selected in MS medium with 400mg/l carboxyl and 40mg/l kanamycin.
