Stylosanthes is an important tropical high-quality legume with high nitrogen content.The application of green manure has this potential effect on soil microbes and the nitrogen cycle in the soil.At present,the effect of applying green manure on the microbiological properties of soils in the manure microsite is still lacking.To this end,this article uses indoor soil columns to study the effect of mixed application of Stylosanthes green manure and phosphate fertilizer on the microbiological properties in the microsite of the fertilizer,in order to clarify the effect of the mixed application of Stylosanthes green fertilizer and phosphate fertilizer on the nitrogen conversion microorganisms in the fertilizer,The study set up the application of low-level green manure GML(including Stylosanthes green manure5 t ha-1),high application of green manure GMH(including Stylosanthes green manure 10 t ha-1),mixed application of green manure GML+MCP(including P2O5 100 kg ha-1+Stylosanthes green fertilizer 5 t ha-1),GMH+MCP(containing P2O5 100 kg ha-1+Stylosanthes green fertilizer 10 t ha-1),single application of phosphate fertilizer MCP(containing P2O5 100 kg ha-1)and control CK(No application of phosphate fertilizer and green fertilizer)six different treatments,the cultivation time is 15 days and 30 days.The main findings are as follows:1.The decomposition and component research of green manure found that 15 days before the application of green manure is the rapid decomposition period of green manure.15 days after the application of green manure,more lignin,cellulose,hemicellulose and other difficult-to-decompose substances remain in the green manure,which is the slow decomposition period of green manure.In terms of fertilizer application,the decomposition rate of GML+MCP and GML is higher than that of GMH+MCP and GMH.The cellulose content after 15 days of cultivation is higher than that after 30 days of cultivation.In terms of mixed application,the cellulose content of green fertilizer GMH and GML after 15 days of cultivation is higher than that of green fertilizer and phosphorus fertilizer mixed with GMH+MCP and GML+MCP,but after 30 days,the cellulose content of GML+MCP and GMH+MCP is higher than The cellulose content of GML and GMH is applied.2.Different fertilizer treatments have different changes on microbial nitrogen content.After 15 days and 30 days of cultivation,the application of green manure can increase the soil microbial nitrogen content,The treatment of MCP with phosphate fertilizer alone had no effect on microbial nitrogen content.GML+MCP treatment had significantly higher changes in microbial nitrogen content than other fertilization treatments.3.In terms of soil bacterial diversity,there was no significant change in bacterial diversity in the fertilization microsite soils of different treatments after 15 days of cultivation.After 30 days of fertilization,the bacterial diversity has changed significantly,and the distance range of different fertilization treatments on the bacterial diversity and impact is also different.The diversity of bacteria began to be affected within 4 mm-20 cm of the microdomains of the manure microsite.Both the application of GMH and the application of GMH+MCP can significantly increase the diversity of bacteria.4.The number of OTU did not increase significantly after 15 days of cultivation.The number of soil bacterial OTUs under the 30-day fertilization treatment was higher than the number of soil OTUs cultured on 15 days,an average increase of 1.27 times.The number of OTU under the treatment of single green fertilizer GML and GMH was higher than that of the mixed application of green fertilizer and phosphorus fertilizer GML+MCP and GMH+MCP.In terms of single application of phosphate fertilizer,there was no significant difference compared with CK.5.Fertilization treatment mainly affected the changes of thick-walled phylum and proteobacteria in the microsite of soil fertilization.The application of high amount of green manure GMH can increase the number of thick-walled phylum,while the application of low amount of green manure GML can reduce the number of proteobacteria.The application of fertilizers reduced the number of actinomycetes.6.The number of nitrogen-fixing bacteria after the application of green manure and mixed phosphate manure increased significantly.The effect of single application of green fertilizer GML and GMH to increase the number of nitrogen fixation is more significant than that of mixed application of green fertilizer GMH+MCP and GML+MCP.
