In the age of post stage of genomics, with the development of gene sequencing ,we get a large of data about it.So, it's important to find the function of the corresponding sequence.Our lab has mapped the boron-high-efficiency gene in rape.And we are looking for boron-high-efficiency gene and related sequence in the Arabidopsis .Microarray is a kind of new technology which make find and analysis special genes in a mass of data possible. Four times backcross and one time of selfcross were carried out.And we get the born high-efficency NILS .the material was applied in the further study.The main result are as followed:1. Establish the born critical concentration: 0.002mg/kg. Boron-high-efficiency Arabidopsis ecotype Lersberg has fertility while boron-low-efficiency Arabidopsis ecotype Columbia has no fertility.2. Screen all the hybrids by both phenotype and SSR MAKERs, and qicken the breeding speed by using of RI lines.3. Acquire the Columbia NIL(nearly isogenitic line),they were applied in the further studyA mutant about born efficiency had been scaned Based on the promoter trap line (our lab get it by the sponsor of 971 items). In this experiment, 3400 individuals of Arabidopsis (boron high efficiency) were selected. The main results are as followed:1. Arabidopsis B4650 exhibited the boron deficiency symptom such as sterility,as boron-low-efficiency individual.2. By GUS analysis, we do not find the corresponding gene express with tissue speciality.It express both in the root and leaf.3. By the improved TAIL PCR, we get the fragment flanked the T-DNA4. BLAST in GenBank and showed it is homologue of sterol-regulatory element binding protein (SREBP) site 2 protease family protein (At5g05740).
