Spermatogonial stem cells—male germline stem cells—is a special kind of somatic stem cells, capable of endlessly self-replication, differentiation of sperm formation, set mitosis, meiosis and cell morphogenesis as a whole; in vitro, SSCs can be induced to differentiate into embryonic stem cell-like cells of pluripotent stem cells. Therefore, the study not only has its SSCs important biological significance, excellent in livestock breeding, genetic modification, as well as the treatment of male infertility, stem cell tissue repair have potential applications. However, self-replicating elaborate SSCs, together dedifferentiation mechanism of differentiation, as well as a prerequisite for the application of biotechnology SSCs is to establish a reliable SSCs in vitro culture system.In order to achieve the above objectives of this long-term study, THY-1 antibody by sorting were successfully isolated from 5-8 days after birth, the testes of male mice of different strains of high purity SSCs, and the establishment of SSCs cell lines, in vitro more than three months.SSCs established cell lines were:expressing CAT / GFP, OCT4 / GFP marker gene, and C57 inbred mice and the genetic background of DBA × C57 mouse SSCs cell lines. In addition, we try to use outside of testicular extracellular matrix scaffold cultured testicular cells, combined with four different culture system, expect to establish spermatogenesis in vitro differentiation system. Preliminary results indicate that the bracket has a support SSCs testicular differentiation capacity, but intensive work remains to be further efforts.
