14 out of 25 strains of biocontrol agent Streptomyces showed chitinolytic activity by biological assay. These 14 strains of biocontrol agent Streptomyces were detected for chitinase gene using PCR primers derived from the conservative sequence of 17 cloned chitinase genes from Streptomyces. A specific 480bp DNA fragment was amplified by PCR from 11 out of the 14 strains of Streptomyces with chitinolytic activity. The Identity of its sequence with S. coelicolor ChiC is up to 90%. It was demonstrated that the PCR-mediated method to detect the chitinase-producing Streptomyces from soil environment is possible and effective, and the primers designed are also qualified.Selected one of the 14 strains-S93, S93 DNA was digested partially with Sau3A I and 2~3kb fragments were collected and inserted into pUC 18, then transformed into DH5 a . Filtering the clone with hybridization in situ, A 1 kb frament clone has been cloned.
