The salt always blocked the agriculture production, and is one of the main reason for the infield desolation. There is three hundred million unit of area in our China. How to solve the salt tolerant not only is the important means to solve the field issue, but also is the one of theissue needing to been solved.Na+/H+ Antiporter is one sort of protein which on the membrane or vacuole membrane, they can transport Na+ retrorsely out of the cell or into the vacuole. Most of the algaes and the salt plants have this kind of protein. The SynNhap gene is cloned from the Synechocystis pcc6803, is the membrane Na+/H+Antiporter. Its open read frame is 1581bp, encoding a peptide of 527 amino acids. Transformanted it to the E.coli Toll4, can recover the TolHs growth ability in the plate with Na+.To make The PCR primer that have adequacy enzyme sit flowed the sequence, ligate with the pGEM-T Easy after PCR. Make sure the sequence by sequencing. To make the Ti plasmid pBI121-Nhap for transformants the tobacco (NC89), at the same time to make the pEmu-mcs-N-Nhap and pWubi.tml-Nhap for transformants the yan wheat 18. We getted 4 transgenic tobaccos, measured the ability of salt tolerance by treatment in 150 mmol/1 NaCl, Found that the transgenic tobaccoes can growth and the control tobaccos can't. We didn't find the transgenic wheat in 347 grains wheat seed.
