Effect of environmental factors on the growth of plant cell and tissue is one of important fields in plant physiology, and that of physical stimulation on genetic substance also is focused on the plant genetic research. Now the research on the effect of sound stimulation on the plant becomes more and more attentive, and it has been found that sound stimulation can greatly influence the growth and development of plant, but the studies about the effect of sound stimulation on the hereditary substance is rare so far. In this thesis, the influence of sound stimulation on the hereditary substance was primarily studied, and the contents and results of this thesis were followed.Firstly, the effect of sound stimulation on protective enzymes of chrysanthemum was explored. In this work, chrysanthemum seedlings were treated by sound wave with a certain intensity (100db) and frequency (1000 Hz) for 3,6,9,12,15 days, respectively, and each day for 60 min. The results showed that the activities of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) increased at varying degrees, and that better promotion for chrysanthemum was obtained under sound wave for 9 days. We also added CHM, a kind of protein synthetic inhibitor, in medium of stimulated group and found the activities of POD and CAT decreased, which implies that the rising activities of protective enzymes were caused by the acceleration of enzyme synthesis in plant cells under sound wave.Secondly, analysis of peroxidase isoenzyme with polyacrylamidedel electrophoresis for was performed in order to investigate the changes of gene expression under sound stimulation. It could be seen from electrophoresis gel that each group had 6 enzyme bands. New enzyme band in POD electrophoretogram wasn't detected for stressed groups. The graph showed that the POD isoenzymes of stimulation groups were dyed deeply than that of control group, and the deepest was the one of T9 group(stimulating for 9 days). The results demonstrated that the activities of POD isoenzymes increased under the sound. Compared with control group, there was no new enzyme band in POD electrophoretogram for stimulation groups. It implies that gene expression in plant cells might have changed under the stimulation of sound wave.Thirdly, the cell cycle of protoplast of control group and stimulated group (stimulating for 9 days) was estimated by flow cytometer. The results showed that the number of cells of stimulated group decreased in G0/G1 phase and increased in S phase. So soundstimulation might accelerate the growth and mitosis of chrysanthemum cells.Fourthly, the contents of DNA, RNA and soluble proteins were measured in this thesis. The content of DNA didn't change evidently and that of RNA and soluble proteins increased at varying degrees. The results demonstrated that expression of gene related to stress response started and the transcription level increased under sound stimulation, and consequently the synthesis of proteins increased. We could infer from the upper results that heredity substance had no variation and only gene expression changes under sound stimulation, and the variation made no inheritance and was just adaptability to environment of plants...
