Plants regulate development transitions in response to a variety of both external and endogenous internal cues. Genetic studies of the transition to flowering in the model plant Arabidopsis thaliana have revealed two major floral promotion pathways. The photoperiod and vernalization pathways, Which are from a quantitative network that regulates the timing of the transition from a vegetative to reproductive phase of development, integrate environmental signals into the flowering determination. The vernalization pathway promotes flowering in response to a long period of cold temperature, usually experienced as plants overwinter. Vernalization has been recognized as a key process in the floral transition for a long time. Vernalization is a quantitative response with increasing periods of low temperature causing progressively earlier flowering until a saturation point is reached. Although all dividing cells are thought to undergo vernalization, only the shoot apical meristem needs to be vernalized to accelerate flowering.In the study, a DNA fragment of Vrn2 gene hi Arabidopsis thaliana was obtained byPCRSpecific primers were syntheticed on the basis of Vrn2 from Genebank. The full-length DNA sequence was 3267 bp,including 5 introns and 6 exons which encoded 503 amino acids. Prosite motif research showes that there may be 2 N-glycosylation site, 6 protein kinase C phosphorylation site, 5 Casein kinase II phosphorylation site, 9 N-myristoylation site, 1 Amidation site. The results of prediction of Protein Localization Sites software indicate that Vrn2 encodes a nuclear-localized protein and PHD-sec shows that the composition of the secondary structure is 37.1% H, 19.1%E and 43.8%L.The results of Southern Blotting indicated that Vrn2 had single copy in the Arabidopsis thaliana genome and Gladiolus Hybridus genome.
