Thuidiaceae, widely distributed in the world, is one of the large families of Hypnobryales. Usually the systematic position of Thuidiaceae is located between Leskeaceae and Amblystegiaceae, however its infra phylogenetic relationship is very confused, therefore in this research project, we try to analyse its phylogenetic relationship using molecular methods to get better results, hi this paper, we used a set of methods suitable for nuclear DNA extraction of mosses, as well, we found an optimal PCR reaction conditions using LA Taq t)NA polymerase. Finally we got 6 species' ITS sequences of Thuidiaceae, and all of them were sequenced directly from PCR products. They are: Abietinella abietina (AJ417494). Anomodon minor (AJ344145), Claopodium aciculum (AJ315968), Thuidium pristocalyx (AJ416443), Thuidium assimile (AJ416442) and Herpetineuron toccoae (AJ315967). Among them, except Abietinella abietina, the other five species' ITS sequences are the first time to be reported. The results of this study are that we used all of our ITS sequences about Thuidiaceae (s.l.) and combined the data downloaded from GenBank databases to establish the phylogenetic tree main of Thuidiaceae (s.l.) and some genera and species of Hypnobryales. Two main paraphyletic clades, one including the genera Anomodon, Herpetineuron and Haplohymenium et al., and the another one including Abietinella, Thuidium and Helodium et al., are recognised. These results, at least, support the morphological study to isolate the subfamily Anomodontoideae from Thuidiaceae (s.l.) and uplift it to be an independent family Anomodontaceae.
