Studies On Isozymes And Their Genetic Effects On Morphological Traits Of Huaihe Carp

The genetic variation of eight isozymic systems (Lactate dehydrogenase (Ldh), Malate dehydrogenase(Mdh), Isocitric dehydrogenase(Idh), Glycero-3-phosphate dehydrogenase (G3pdh), Malatase (Me), Esterase (Est), Alcohol dehydrogenase (Adh), Glutamate dehydrogenase(Gdh)) encoded approximately by 25 presumptive loci in 46 samples of huaihe carp (Cyprinus carpio var. huaihenensis) were detected through discontinuous vertical plate polyacrylamide gel electrophoresis (PAGE). Polymorphisms were observed at six loci (Ldh-C, Idh-A, Est-1, Adh-A, Gdh-1, 2) , The observed frequencies of genotypes at five (Except Idh-A) polymorphic loci were all in accord with the Hardy-Weinberg. It can be speculated that the population of huaihe carp was a relative independent stock by the studied results of chi-square. The results showed that the isozymes loci of huaihe carp possess obvious polymorphism. The proportion of polymorphic loci and the mean average heterozygosity of the population were 24%and 0.086,respectively. Polymorphism of serum Transferring (Tf), Est, Ldh by PAGE in 127 samples and the morpholigical traits at 8,13,20 and 24 months of age of huaihe carp were detected, respectively, furthermore, the genetic effect of the polymorphism of serum protein on the morphological traits were analyzed. The results showed that Tf, Est-1 and Ldh-C were all polymorphic. The Tf locus had three codominant alleles (TfA, TfB, TfC), Their frequencies were 0.7204,0.2402and 0.0394 respectively. The gene frequency for T^ was the highest. There were five phenotypes (TfAA, TfAB, TfBB, TfAC and TfAC ) and their frequencies were 0.5669,0.2598,0.0945,0.0472and 0.0316 respectively. The frequency for TfAA was the highest. The genotypes distribution of Tf was in accord with Hardy-Weinberg equilibrium. The index of genetic polymorphism at the Tf locus was 0.4218.Body weight at 20 and 24 months of age with TfAA were significantly higher than those with other genotypes. The Est-1 locus had two codominant alleles (Est-lAand Est-1B) .Their frequencieswere 0.6024 and 0.3976 respectively, The gene frequency for Est-lA was higher. Three phenotypes (Est-lAA, Est-lAB and Est-lBB) were detected. Their frequencies were 0.3543,0.4961 and 0.1496, respectively. The genotypes distribution of the Est-1 locus was in accord with Hardy-Weinberg equilibrium. The frequency for Est-1AB was highest. The index of genetic polymorphism at the Est-1 locus was 0.4790. Body weights at all phases with genotype Est-1AB were the highest and were significantlyhigher than those with Est-1BB.The Ldh-C locus had three codominant alleles (Ldh-CA,Ldh-CBandLdh-Cc) .Their gene frequencies were 0.0866% 0.6063and 0.3071 respectively. The frequency for Tf3 was highest. There were four genotypes (Ldh-CA8, Ldh-CBB, Ldh-CBCand Ldh-Ccc ) at the Ldh-C locus and their frequencies were 0.1732, 0.3701, 0.2992 and 0.1575 respectively, The frequency for Ldh-C was highest The genotypes of Ldh-C^ and Ldh-CAC were not detected, The genotypes distribution of Ldh-C deviated from Hardy-Weinberg equilibrium. The index of genetic polymorphism at the Ldh-C locus was 0.4694.Body weight at 13, 20and 24 months of age with genotype Ldh-CBC were significantly higher than those with genotype Ldh-CAB, but they were no significant difference with genotypes Ldh-CBB and Ldh-Ccc.Effect of different Tf,Est-l,Ldh-C genotypes on body type traits also were analyzed by least squares, The results showed that effect of three loci on body length, tailstalk length decreased gradually, while the effect of three loci on body height, body width increased little by little. But the effect of three loci on head length and other morphological traits were no obvious regularity. Therefore, it was thought that the traits of body length,tailstalk length should be selected earlier and the traits of body length, body width should be selected later while huaihe carp are growing up.
