THE STUDY OF EXTRACTION TECHNIQUES OFISOFLAVONE FROM SOYBEAN RESIDUEIt is one of secondary metabolites formed in the growth of soybean. The principal chemical forms of isoflavones in soybean are conjugates. It has many important biological activities and is rich in soybeans and soybean products, It may be applied in several fields, such as in drugs, heahh foods and so on.This study was based on the former working foundation of our soybean research group. The author aimed at technological process of extracting soy crude isoflavones using soy residue as raw material, of hydrolysis extraction and of purifying isoflavones by macroporous adsorption resin method. The author also found that the method of measuring the content of isoflavones by UV spectroscopic analysis and the content of genistein using pure genistein as standard by RP-HPLC, respectively. The results showed that:I.The best conditions for the extraction techniques of isoflavones were: M alcohol, reflux extracting 3 times, 3 hours per time, the ratio of solvent and sample 3:1. The ration of extracting 32.73%,isoflavorie content 22.24%.2.The excellent technological process to acquire genistein by hydrolysis method was: O.lmol/L hydrochloric acid, 3 hours, temperature 400C, the ratio of hydrochloric acid and sample 5 1. Genistein content 5.95 %,the ration of break5.25%.3.The optimal technological process to purify isoflavones by macroporous adsorption resin method was: 50% alcohol, the ration of solvents and samples 230 1, the retention time 2 hours.4.To measure the content of genistein in soybean extracting material, RPHPLC method was used. The separating process was performed on Hypersil BDS C18 Colum (250mmX4.6mm, 5~trn), with a mobile phase of CH3CN-0.OO3mol/L44Na2HPO4, flow rate of 1.0 mi/mm, and detecting wavelength at 259 nm.RSD 0.056%,the recovery for genistein 99.67?.056)%.This study provides solid theoretical foundation for the development and utilization of soy isoflavones. It has strong guiding significance in practical working.
