Umbilicariaceae is a kind of Lichen which lived in special enviroments. It usually lived on the surface of rocks in high altitude or circumpolar latitude areas. Some species of Umbilicariaceae Shows the bioactivity of antitumor, antimicrobial, anti-HIV and enzyme inhibition. Reaserch on the lichenized fungi of Umbilicariaceae has never been reported before. The author studied the activity of lichenized fungi Umbilicaria muehlenbergii with the expect that it will afford the reliable data support on the exploit of Lichenized forming fungi of Umbilicariaceae.A new methord for isolation and culture of Lichenized fungi was used and yeast form of lichenized fungi Umbilicaria muehlenbergii was obtained. The growth rate of 71.5% and contamination rate of 7.14 was much better than that of 60.71% and 21.43 of Yamamoto methord.The ITS sequence of yeast form lichenized fungi Umbilicaria muehlenbergii had been comparied with lichen thallus Umbilicairia muehlenbergii . The results shows the similarity was 99%.The orthogonal test has been done to study the ferment condition of yeast form lichenized fungi Umbilicaria muehlengbergii. The result shows that MS culture medium, 25℃,carbon source mannitiol was best ferment condition.The E.A extract, aqueous extract yeast cell and solid ferment extract of ferment products was used to study the antimicrorial activity. According to the result: Almost no samples shows activities against staphylococcus aureus, enterococcus faecium and helicobacter pylori.The anti-tumor activity of the aqueous extract of ferment product had been tested and no activity against AGS cancer cells represented according to the result.Anti-oxidant activity has been tested by DPPH free radical scavenging rate, anti linoleic acid overoxidation rate and reducing power. According to the experiment results, the DPPH free radical scavenging rate reached 65.18% at the concertration of 330ppm,and the anti overoxidant linoleic acid rate 74.45%,reducing power 0.88 perspectively.The convertion condition from yeast form to hypha form of lichenized fungi Umblicaria muehlenbergii has been studied. According to this study, hypha formed fungi has been induced under changed culture tempature. The stavadion condition also takes some part of the convert progess.
