Analysis Of Ubiquitin-proteasome System About Senility Of Alzheimer Disease And Parkinson Disease With The Kidney Deficiency Syndrome

Objective: To analyze dysfunction biological mechanism of senility-ubiquitin proteasome system about the kidney deficiency syndrome of Alzheimer Disease(AD)and Parkinson Disease(PD)that the different diseases with same syndrome..Methods: 1.Proposed scientific hypothesis and demonstrate the contents and theoretical basis of scientific hypothesis by theory analysis.2.Observe behavior of the AD& PD animal model that combined relational diseases and syndromes,qualitative and quantitative change of apoptosis,HPA and senility anti-oxidant function,UPS protein and gene expression and intervention effect of Dihuangyinzi(a kind of soup of Chinese medicine)of AD & PD with light microscope,enzyme-linked immunosorbent assay,Td Tmediated d UTP nick end labeling,Western-blot,RT-PCR to demonstrate the biological mechanism assumption about the theory of different diseases have the same syndrome in AD & PD.Results:1.Theoretical research.Traditional Chinese medical thinks that kidney vacuity and essence depletion is the reason of AD & PD,which can be concluded as the same syndrome in different disease.Western medicine thinks that UPS dysfunction caused by senility is the pathomechanism both for AD and PD.Traditional Chinese and western medicine define senility as physiology kidney deficiency.The author attempts to demonstrate the assumption that UPS dysfunction caused by senility is the syndrome both AD and PD.2.Experiment research.(1)The memory of the AD rats model is damaged roughly and nerve cells in hippocampus apoptosis increase which is to say that AD model is made successfully.Aniti-oxidant ability of rats decreases and HPA axis hyperfunction through sera diagnosis show that kidney vacuity model is made.(2)Equilibrium of the PD rats and initiative motion weaken,and aniti-oxidant ability of rats decreases,and nerve cells apoptosis in substantiae nigrae increase,which show that PD model is completed.Aniti-oxidant ability of rats decreases and HPA axis hyperfunction through sera diagnosis show that kidney vacuity model is made successfully.(3)Intervention of Dihuangyinzi will improve memory of rats,decrease nerve cells apoptosis in hippocampus,enhance anti-oxidant ability of rats and improve HPA axis hyperfunction,and it is not effective for equilibrium of rats and initiative motion,however,it will decrease nerve cells apoptosis in substantiae nigrae,enhance aniti-oxidant ability and improve HPA axis hyperfunction.(4)The test results of ubiquitin-proteasome system are that m RNA and protein content test results are consistent,and PUB of rats in AD model,but it declines after watering Dihuangyinzi.?-syn and PUB increase in PD model,but it declines after watering Dihuangyinzi.Dihuangyinzi nourishes kidney yin and kidney yang.It is true that senility and UPS dysfunction are biological mechanism which causes AD&PD.Conclusions: It is shown that it exists in intervention effect of Dihuangyinzi kidney vacuity of AD & PD to enhance antioxidant capacity,regulate HPA axis hyperfunction,recover UPS dysfunction and decrease nerve cells apoptosis to analyze the test results.So,it prove “Senility-dysfunction of ubiquitin proteasome system is a part of biological mechanism about the kidney deficiency syndrome of AD and PD that the different diseases with same syndrome ”is correct.
