Objective To observe the relativity and diversity of balanced constitution(healthy persons),phlegm-dampness constitution and yang-deficiency constitution on the aspect of gut microbiota;To study the effect of regulating phlegm-dampness constitution and remolding gut microbiota by Herb-partitioned Moxibustion on Navel firstly;To explore and analyze the mechanism of reconstruction of gut microbiota and regulation of phlegm-dampness constitution by herb-partitioned moxibustion on navel by means of 16 s function prediction.Methods1 60 persons with phlegm-dampness constitution,30 yang-deficiency constitution and 30 healthy persons were recruited according to the including criterion,which were totally 120 persons.2 The total bacterial DNA were extracted from the three groups of samples including 30yang-deficiency constitution,30 healthy persons and 30 persons which were extracted randomly from the 60 persons with phlegm-dampness constitution to analysis the difference of constitution and abundance of three groups by analyzing the 16 S r DNA sequencing.3 60 persons with phlegm-dampness constitution were divided into two groups randomly.The QJ group(observation group)and control group were 30 persons respectively.The QJ group received Herb-partitioned Moxibustion on Navel and regimen guide while the control group only received regimen guide in 8 weeks.4 The observation indices were the constitution and abundance of gut microbiota,transformation score of phlegm-dampness constitution,BMI,waist circumference and so on.5 The mechanism of structural alteration of gut microbiota and regulation of phlegm-dampness constitution by herb-partitioned moxibustion on navel were analyzed in the way of 16 s function prediction which mainly utilize the software of PICRUSt to analyze COG function classification from database of Egg NOG and acquire pathway map from database of KEGG.Results1 On the classification level of OTU,partial least squares-discriminant analysis(PLS-DA)showed that there was a significant difference in the composition of intestinal flora among the phlegm-dampness constitution,the balanced constitution and the yang-deficiency constitution.On the classification level of genus,the abundance of Roseburia were the most significant difference among three groups(P < 0.05)by Kruskal-Wallis rank test,which were the lower in yang-deficiency constitution persons than it in healthy persons and were the lowest in the phlegm-dampness constitution persons.2 The main change of gut microbiota of QJ group and control group manifest in the genus level that the abundance of [Eubacterium]_hallii_group increased significantly in QJ group(P<0.01),while the abundance of Blautia decreased distinctly in control group(P<0.05)by Wilcoxon rank test.The QJ group and the control group both can decrease the indexes of transformation score of phlegm-dampness constitution and waist circumference,while the QJ group were evidently superior to the control group(P<0.01).Furthermore,the indexes of BMI(P<0.01)and hip circumference(P<0.05)of QJ group had remarkably decreased.3 The major functions in QJ group by the analysis of COG function classification are the function of carbohydrate transport and metabolism,amino acid transport and metabolism,transcription,general function prediction only,replication,recombination and repair,translation,ribosomal structure and biogenesis.The main pathway of structural alteration of gut microbiota and regulation of phlegm-dampness constitution by herb-partitioned moxibustion on navel are the ways of glycolysis and gluconeogenesis,starch and sucrose metabolism,pentose phosphate pathway,carbon fixation in photosynthetic organisms,pyruvate metabolism,propanoate metabolism,citrate cycle.Conclusion1 There are difference in abundance and species of intestinal flora among the phlegm-dampness constitution,the yang-deficiency constitution and the balanced constitution,especially manifested in the difference on the aspect of ratio of relative abundance of Roseburia,which in the phlegm-dampness constitution was the lowest.2 Herb-partitioned moxibustion on navel not only can decrease the indexes of transformation score of phlegm-dampness constitution and waist circumference,and BMI,but also can significantly increase the abundance of [Eubacterium]_hallii_group,which indicate that herb-partitioned moxibustion on navel have the function of reconstruction of gut microbiota and regulation of phlegm-dampness constitution.3 The major functions in QJ group are the function of carbohydrate transport and metabolism,amino acid transport and metabolism.Furthermore,glycolysis and gluconeogenesis are the primary pathway of reconstruction of gut microbiota and regulation of phlegm-dampness constitution by herb-partitioned moxibustion on navel.
