There are 1012-1014 microorganisms in the intestines of adults.Therefore,the gut microbiota is also known as another major"organ"of the human body.The gut microbiota can act on the central nervous system through the gut-brain axis,affecting the metabolism and behavior of the intestinal barrier system.Changes in the human diet and life lead to changes in intestinal metabolites,which affect the body's immune barrier and nervous system,and ultimately affect health.Lacticaseibacillus rhamnosus Probio-M9(Probio-M9)is a strain with potential probiotic effects isolated from breast milk.Ninety adult volunteers were selected for a 6-week intervention in a randomized,double-blind,placebo-controlled trial.Metagenomic sequencing technology to were used to analyze the gut microbiota of volunteers.Together with results from fecal metabolome,blood metabolome,blood immune indicators,and health status survey to comprehensively evaluate the impact of Probio-M9 on adults'quality of life.The results of the study are as follows:(1)Probio-M9 can be colonized in the host's gastrointestinal tract and increase the alpha diversity of adult intestinal microbiota(P<0.05).Probio-M9 regulates host intestinal microbiota via increasing the contents of Bifidobacterium adolescentis,Bifidobacterium longum,Bifidobacterium pseudocatenulatum,Dorea longicatena,Eubacterium rectum and Lacticaseibacillus rhamnosus in the intestine(P<0.05),which has a positive effect on the health of the body.(2)Probio-M9 significantly increases the content of short-chain fatty acids such as propionic acid,butyric acid and isobutyric acid in the intestines of volunteers(P<0.05).Up-regulates the metabolic pathways of arachidonic acid,alpha-linolenic acid and linoleic acid,and increases the beneficial products of arachidonic acid,leukotriene B4,prostaglandin E2 and linoleic acid in the pathway are increased after Probio-M9intervention showing a positive effect on the body's health.At the same time,the same trend is shown in blood metabolism.(3)Probio-M9 regulates the blood levels of eosinophil chemokines,interleukin-1?,interleukin-8,monocyte chemotactic protein-1 and RANTES chemokines in the normal range to maintain a stable level of immune cells,and participate in immune regulation of the body.Thereby maintaining the balance of the body's physiological state.(4)Probio-M9 increases beneficial metabolites such as arachidonic acid,docosahexaenoic acid(DHA),vitamin B2,dopamine,and tryptophan(P<0.05)in the intestine by regulating the intestinal tract,which makes adulthood Human gastrointestinal and upper respiratory tract infections a,mental and psychological symptoms were improved(P<0.05),thereby improving the quality of life of volunteers.In summary,our research indicates that intaking Probio-M9 increases the alpha diversity of the host's gut microbes,increase the content of beneficial bacteria such as Bifidobacterium adolescentis and Bifidobacterium longum,and promotes the generation of beneficial metabolites such as short-chain fatty acids,dopamine,and tryptophan in the host's intestinal tract.This may indirectly affect the host's blood metabolism pathway,play a positive role in the balance of the gut microbiota and immune regulation of volunteers,and ultimately improve adults'quality of life comprehensively.The results of this study provide evidence-based evidence for the probiotic effects of Probio-M9 and theoretical support for the development of probiotic supplements suitable for adults.
