Objectives: 1)To analyze the expression of FUT3 in clear cell renal cell carcinoma.2)To study whether the expression of FUT3 is an independent risk factor for the prognosis of patients with ccRCC.3)To evaluate the prognostic predictive value of FUT3 expression in ccRCC,and try to provide a new choice for clinical prognosis prediction and stratified management of patients.Type of study: A retrospective cohort study consisting of 406 patients with histo-pathologically proven ccRCC followed up for 74 months.Research methods and contents: The expression of FUT3 was analyzed by immunehistochemical staining of tissue microarray and semi-quantitative scoring.With the help of statistical and mapping software,the correlation between the expression of FUT3 and the clinic-pathological characteristics of patients are assessed and the survival curve was made.Cox proportional hazard regression was used to find independent risk factors related to prognosis;Nomogram was made to establish prognostic prediction system.Concordance index and AIC value were calculated to evaluate the predictive ability of established prognostic system integrated with FUT3 expression.Results: Patients with ccRCC with high expression of FUT3 in cancer tissue were inferior to those with low expression of FUT3 in both OS(p<0.05)and RFS(p<0.05).FUT3 is an independent risk factor of both OS(HR=1.907 95%CI=1.104-3.296)and RFS(HR=1.87995%CI=1.101-3.206)in patients with ccRCC.Besides with high expression of FUT3,tumor size,pT3-4,lymph node metastasis,distant metastasis,microscopic coagulativenecrosis,sarcomatoid change,lympho-vascular invasion is also independent risk factor of OS in patients with ccRCC.On the other hand,Tumor size,pT3-4,microscopic Fuhrman nuclear grade 4,coagulative necrosis,sarcomatoid change,lympho-vascular invasion is also independent risk factor of RFS in patients with ccRCC.Conclusion: High expression of FUT3 in tumor tissue is an independent risk factor for poor prognosis of ccRCC.Combining with FUT3,the prognostic prediction nomogram is built and is easy to use and well-visualized.The classical establish prognostic model of renal cell carcinoma with integration of FUT3 can predict the prognosis of patients more accurately.
