Research purposes:Data mining on the medical records of Professor Zhong-ying Zhou in the treatment of chronic colitis.summarizing and inheriting clinical experience and academic ideas to guide clinical practice.At the same time,a network pharmacological analysis of the common basic prescriptions of Professor Zhong-ying Zhou for the treatment of chronic colitis was carried out to explore its mechanism of action.Research method:(1)Collect the outpatient medical records of Professor Zhong-ying Zhou in the treatment of chronic colitis from October 1989 to December 2014.According to the inclusion criteria and exclusion criteria,176 cases of 617 medical records were selected as the object of study,and the database was established.By using frequency statistics,association rules,cluster analysis,and factor analysis methods to systematically mine the four diagnostic information,pathogenesis,and prescriptions in medical records.Combining medical case study and Professor Zhong-ying's Zhou related works,exploring the mining results,and extracting clinical experience and academic ideas from it.(2)Combined with text reading and based on the results of data mining,appropriate selection of the basic prescriptions of Professor Zhong-ying Zhou to treat chronic colitis for network pharmacology research.Using TCMSP,a traditional Chinese medicine system pharmacological analysis platform,to select active ingredients and regulatory targets,Screening of targets related to chronic colitis through the GeneCard database,and constructing a basic formula-target network diagram;Based on the STRING database,construct an interaction network diagram of the target for treating chronic colitis,and determine the core target based on the parameters;Using Bioconductor database and R software to analyze the Gene Ontology(GO)enrichment analysis and KEGG pathway enrichment analysis of the intersection target of chronic colitis and basic side.The results are analyzed and the mechanism of the basic side is discussed.Research results:(1)General situation:Among the 176 patients with chronic colitis,there was no significant difference in male to female ratio,and there were slightly more women than men.There were 82 males(46.59%)with 276 consultations(44.73%)and 94 females(53.41%)with 341 consultations(55.27%).(2)Four diagnostic information:in all medical cases,the main clinical symptoms were abdominal pain(42.30%),loose stool(38.57%),borborygmus(28.36%),mucus in stool(26.09%),abdominal distension(25.61%),frequent stool(25.28%),break wind(22.53%),dry mouth(17.34%),anal weight(8.91%)and dry stool(5.19%).The main tongue images are thin yellow greasy(38.74%),dark tongue proper(23.82%),deep red tongue(20.91%),yellowish(18.80%),dark red(17.67%),thin yellow(13.61%),greasy and yellow fur on the tongue(13.45%).The main pulse is thin(66.77%),string(51.54%),slippery(27.39%),small(18.48%).(3)Pathogenesis:among the 617 medical case,compound pathogenesis is the main,single pathogenesis only accounts for 77 times,compound accounts for 540 times,accounting for 87.5%.The main pathogenesis frequency from high to low is the discord of the spleen and the liver(24.31%),hot and humid bowels(23.01%),imbalance of fu-qi(19.29%),the Liver restricting the Spleen(16.21%),spleen deficiency and stomach weakness(12.97%),wetness-heat stagnated in the middle warmer(10.21%).The main diseases were spleen(78.28%),liver(58.35%),intestine(53.81%),stomach(33.87%),kidney(8.91%),liver+spleen(50.08%),spleen+kidney(8.43%).The main diseases are heat(56.89%),dampness(54.62%),Qi stagnation(31.12%),qi multiplication(20.75%),cold(10.37%),deficiency(50.73%),damp-heat(49.11%),cold and heat(9.40%).(4)Prescription medication:so in the consultation,there are 32 drugs with a frequency of more than 100.The top 10 flavors were Coptis chinensis(79.90%),atractylodes rhizome(71.47%),White Peony Root(69.37%),Evodia rutaecarpa(63.21%),tangerine peel(55.92%),licorice(47.33%),Radix Aucklandiae(43.76%),roasted ginger(43.44%),lanceolata(40.36%)and radix saposhnikoviae(38.57%).Common drug combinations such as:Coptis chinensis+Evodia rutaecarpa+White Peony Root;medicated leaven+Hawthorn+Gallus gallus domesticus;atractylodes rhizome+Peony+radix saposhnikoviae+tangerine peel;Glycyrrhiza+lanceolata+Roasted Ginger+Chinese Yam;Schisandra chinensis+Nutmeg+Psoralea Fruit+Cinnamon+aconite;Chinese waxgourd seed+ Coix seed+Toon root white bark+Balloonflower+Patrinia villosa;Typha orientalis+Fafces Trogopterori+Fructus Toosendan+Aspongopus+Rhizoma Corydalis;citrus aurantium+Fructus Trichosanthis+areca catechu+radish seed+ Rhubarb+ Prunus Tomentosa Thunb.(5)Results of Network Pharmacology Research:After screening,71 active components in the basic prescriptions(Atractylodes,radix saposhnikoviae,White Peony Root,Tangerine Peel,Coptis chinensis,Evodia Rutaecarpa,rose,chinese waxgourd seed,Coix seed,patrina villosa juss,Balloonflower)can act on 165 targets of chronic colitis,the core targets are AKT1,IL6,PTGS2,etc.,and there are 145 entries in GO enrichment analysis and 163pathways in KEGG enrichment analysis.Research conclusion:(1)Inheritance research:The etiology of chronic colitis is complex,spleen deficiency is often the root cause of the disease.The discord of the spleen and the liver and hot and humid bowels are the common pathogenesis.The disease is located in the intestine,which is related to spleen,stomach and liver,and is related to the kidney for a long time,and the pathological factors are mainly damp-heat,deficiency,Qi stagnation and blood stasis for a long time.The treatment of this disease is mainly for the treatment of the spleen,liver and intestines.Treatment of spleen includes transportation of spleen,nourishing spleen,warming spleen,etc.The treatment of the liver is like suppressing the liver and supporting the spleen,and attention should be paid to the difference of damp-heat,dryness-heat and stasis-heat in the intestine.The treatment of this disease also needs to pay attention to adjusting the human body air movement,to avoid abuse of astringent patients with astringent therapy.In addition,attention should be paid to the symptomatic treatment of mucous stool and severe abdominal pain.(2)Discussion on Mechanism:The 71 active ingredients in the base formula have biological functions such as anti-inflammatory,antibacterial,repair of intestinal mucosa,and can cross-regulate 165 targets related to chronic colitis.It exerts biological functions by affecting the activity of nuclear receptors,the activity of transcription factors,the activity of steroid hormone receptors,the binding of ubiquitin-like protein ligase,the heterodimerization activity of proteins,the binding of transcription cofactors,etc.Regulation of multiple signal pathways including the PI3K/AKT signal pathway,IL-17 signal pathway,and TNF signal pathway to achieve clinical efficacy in relieving intestinal inflammation.
