Purpose: To investigate the effects of acupuncture on blood lipids,hemorheology,inflammatory factor levels,oxidative stress markers and lipid deposition in peritoneal macrophages in rabbits with atherosclerosis,and to explore the treatment of atherosclerosis by acupuncture.The mechanism of action provides an experimental basis for acupuncture treatment of atherosclerosis.Material and method: Twenty-six male New Zealand rabbits(2-3 months old,2-2.5 kg/only)were randomly fed into a blank group and a model group,and 7 of them were blank groups.The blank group was fed with standard rabbit feed,and after 4 weeks of high-fat feeding,the carotid balloon injury was performed,and high-fat feeding was continued after the operation.Four weeks later,one rabbit was randomly selected from the blank group and the model.The pathological changes of the carotid artery in rabbits were observed by HE staining,and the modeling was confirmed.Then,the remaining 18 rabbits were randomly divided into model control group,electroacupuncture treatment group(AS model + electroacupuncture),and positive control group(AS model + atorvastatin calcium tablets),with 6 rats in each group.The blank group was fed with standard rabbit feed without any intervention factors;the model group received no treatment and continued high-fat feeding;the electroacupuncture group was treated with electroacupuncture at Neiguan,Zusanli and Guanyuan points,and the drug group was given atorvastatin.The suspension of calcium tablets was intragastrically administered.After the treatment was finished,the blood lipid content in the blood was detected by chemical method;the blood rheology index was detected by automatic blood rheometer;the serum CRP and TNF-? were determined by enzyme-linked immunosorbent assay.The content of IL-6 and ox-LDL was determined by thiobarbituric acid method.The activity of SOD was determined by xanthine oxidase method.The expressions of CD36 and SR-A1 in peritoneal macrophages were detected by immunohistochemistry.The lipid deposition in peritoneal macrophages was observed by oil red O staining.Results:1.HE staining resultModel evaluation: The carotid artery was observed under the microscope.The carotid artery structure was normal in the blank control group,the endothelium was intact,no inflammatory cell infiltration and foam cell formation were observed.The intima of the model was thickened and the infiltration of the inflammatory cells was observed under the intima.A large number of foam cells gathered under the intima,suggesting that the modeling is successful.Treatment results: Compared with the blank group,the intimal intima of the model control group was significantly thickened and the structure was impaired,and inflammatory cell infiltration,foam cell aggregation,and atheromatous plaque formation were observed.Compared with the model control group,the electroacupuncture treatment group and the positive control group were compared.The area of atherosclerotic plaques was significantly reduced.2.Blood lipid levelsCompared with the blank group,the model control group TG,CHO,LDL-C were significantly increased(p<0.01);HDL-C was significantly lower(p<0.01),compared with the model control group,electroacupuncture treatment group,positive control group The contents of CHO,TG,LDL-C were decreased(p<0.01),and HDL-C was significantly increased(p<0.01).3.Hemorheological indicatorsCompared with the blank group,the model control group showed complete blood viscosity(high cut,medium cut,low cut),capillary plasma viscosity,whole blood reduced viscosity(high cut,medium cut,low cut),red blood cell aggregation index,and hematocrit were obvious.Increased(p<0.01).Compared with the model control group,the electroacupuncture treatment group,the positive control group had whole blood viscosity(high cut,medium cut,low cut),capillary plasma viscosity,whole blood reduced viscosity(high cut,medium cut,low cut),erythrocyte aggregation index,hematocrit was significantly reduced(p<0.01).4.Inflammatory factor levelsCompared with the blank group,the CRP,TNF-?,and IL-6 in the model control group were significantly increased(P<0.01).Compared with the model control group,the electroacupuncture treatment group and the positive control group were CRP,TNF-?,and IL-6.The content was decreased(P<0.05 or P<0.01).5.Oxidative stress marker levelsCompared with the blank group,the SOD content of the model control group was significantly decreased(P<0.01),and the ox-LDL and MDA contents were increased(P<0.01).Compared with the model group,the SOD content of the electroacupuncture treatment group and the positive control group increased.High(P<0.01),ox-LDL,MDA content decreased(P<0.01).6.Expression of CD36 and SR-A1 in peritoneal macrophagesCompared with the blank group,the expressions of CD36 and SR-A1 protein in peritoneal macrophages of the model control group were significantly increased(P<0.05);compared with the model control group,the expressions of CD36 and SR-A1 protein in peritoneal macrophages of the electro-acupuncture treatment group and the positive control group were significantly decreased(P<0.05).7.Lipid deposition in peritoneal macrophagesCompared with the blank group,the lipid droplets in the macrophage of the model control group increased significantly;compared with the model control group,the lipid droplets in the macrophages of the positive control group were significantly reduced compared with the model control group.Conclusion:1.Acupuncture can effectively regulate blood lipid levels and blood rheology indexes in rabbits with atherosclerosis,and has the effect of improving blood lipid metabolism and blood rheology.2.Acupuncture can effectively regulate inflammatory factor levels and oxidative stress markers in rabbits with atherosclerosis,and has certain anti-inflammatory and anti-oxidative effects.3.Acupuncture can reduce the expression of CD36 and SR-A1 in peritoneal macrophages of atherosclerotic rabbits,reduce lipid accumulation in macrophages,inhibit foam cell formation,and thus treat atherosclerosis.
