Experimental Study On The Mechanism Of Shencao Tongmai Granules' Intervention In Chronic Heart Failure Rats

Purpose:Through combination of disease and syndrome principle,the method to create the Rat model reflecting qi deficiency leading to blood stasis syndrome(QDLTBSS)of chronic cardiac failure was also researched.The curative effect of Sen Cao Tongmai Particle(SCTMP)treating chronic cardiac failure rat of QDLTBSS was researched,offer evidence based medicine to traditional Chinese medicine.The main object of this research is to explore the chronic heart failure heart spleen related mechanisms and to observe the expression of 5-HT,IGF-1,GLUT4,VCAM-1 and AQP2 in Myocardial tissue,kidney tissue and aorta tissue.Material and method:1.Randomly selected the 10 rats from 100 SD rats that were not treated with any treatment were blank control group.The 90 rats were treated with adaptive feeding for 1 week,and were made moulded,disease index such as cardiac function,BNP were detected.Further,syndrome information including heart rate,breathe,urine volume and so on were detected.2.The successful rats were divided into the model group,formulae of invigorating Qi and promote blood circulation and the lisinopli group according to the random number table method.4 weeks later,death rate,cardiac function,BNP,heart rate,respitary rate were detected.Further,syndrome information including heart rate,breathe and urine volume were detected.3.Determing the concentration of 5-HT,IGF-1,GLUT4,VCAM-1and AQP2 by ELISA,Western blot,immunohistochemical method and real time quantitative PCR among TCM group,Western group and control group,model group.Comparing the structure changes of histomorphology and cardiomyocytes of four groups under light microscope.Results:1.The index reflecting the disease and syndrome reflected the model of CHF ratswas sucessful.2.The index reflecting the disease and syndrome were improved by drug.3.5-HT,IGF-1,GLUT4,VCAM-1 and AQP2 were improved by drug.Conclusion:Ligation rat left anterior descending coronary artery,underfeed after operation,load exercise was an appropriate method to create rat model reflecting QDLTBSS of chronic cardiac failure.Beside SCTMP could improve heart function,it could improve syndrome index and BNP,5-HT,IGF-1,GLUT4,VCAM-1 and AQP2 may be the mainmechanism for the treatment of heart failure.SCTMP might be an assistant drug to cure chronic heart failure.
