Objective: To investigate the relationship between the recurrence of colorectal adenoma and the changes of intestinal microflora.Methods: From November 2009 to September 2014,patients undergoing colorectal adenocarcinoma resection were enrolled in the Department of Gastroenterology,Ren ji Hospital,Shanghai Jiaotong University.After 4 years of follow-up,40 patients were selected and divided into recurrence group and recurrence-free group.Then their fecal samples were collected preoperatively and extracted the DNA from them.The Miseq high-throughput sequencing of the 16 S rRNA gene V4-V5 region was used to analyze the biotic diversity,diversity and diversity of the intestinal flora.Then real-time quantitative PCR was used to validation the results.Results: The population structure of intestinal flora was similar in both groups.The Firmicutes,Bacteroidetes and Proteobacteria were the most important microflora in the patients with colorectal adenoma.On phylum level,Fusobacteria had significant differences in the recurrence group and the recurrence-free group(P = 0.00849).There were more strains of the Fusobacteria in the recurrent group and other microflora had no significant difference.On genus level,these florae in recurrence group were more than recurrence-free group: no-rank genus in Fusobacteria,Lachnoanaerobaculum and Parvimonas.these florae in recurrence group were less than recurrence-free group: Eubacterium,Sutterella,Christensenella and Aeromonas.The above results were confirmed in the PCR assay,and the differences between the two flora could be directly detected by PCR.Conclusion: In colorectal adenoma patients,who has the higher abundance of Fusobacteria,and lower abundance of Eubacterium,has higher risk of recurrence.
