Objective:1.To explore the feasibility of using endometrium derived from menstrual blood in studying pathogenesis of endometriosis;2.To demonstrate the attachment-aggression-angiogenesis process in Determinant Of Uterine Eutopic Endometrium;3.Further explore the possibility of menstrual-blood-derived markers as new noninvasive approach for endometriosis diagnosis;Methods:1.Menstrual blood collected by menstrual cup were filtered for primary cell culture or cryopreserved endometrium.2.TMT labeling for high performance liquid chromatography(HPLC)and mass spectrometry were used for quantitative proteomics of menstrual blood endometrium;3.Bioinformatics analysis of differential proteins in the endometriosis group compared to the control;4.Immunohistochemical staining were done for further verification of menstrual endometrium and cells;Results:1.The amount of menstrual blood collected from the two groups has a significant difference.There was no difference between the weight of extracted endometrial tissue or protein of two groups;2.A total of 4243 proteins were identified,and 1357 differentially expressed proteins were obtained with the E/C ratio of 1.2 times as the change threshold;3.A large group among the identified differential expressed proteins in the endometriosis group,play important roles in cell adhesion,invasion and angiogenesis,including integrins super family and its ligands,CD44,Focal adhesion kinase 1,anchor proteins of focal adhesion,collagens,Hyaluronans,matrix metalloproteinases family and its inhibitors,angiopoietin and other proteins;Conclusions:1.Menstrual-blood-derived endometrium is a good source of specimens to study the pathogenesis of endometriosis;2.Further confirmation of Determinant Of Uterine Eutopic Endometrium;3.It has important guiding significance for the screening,diagnosis and treatment of endometriosis;...
