Through researching and reviewing Massage Technique of pressing's literature,both ancient theories and modern clinical application and experimental exploration have been used to analyze “After Pressing,It Heat Up”'s basic connotation.Based on a large number of clinical practice in pre-Qin period,the theoretical framework of “After Pressing,It Heat Up” has been established in Huangdi Neijing,further applied to clinical practice in Wei Jin Northern and Southern Dynasties,sublimed with accumulated clinical practice in Sui and Tang Dynasties,theoretical mechanism further excavated in Song Jin Yuan Period,rapidly developed the theory and clinical practice during the Ming and Qing Dynasties.This theory comes from practice,which has stood up the test of practice and theoretical improvement,is a massage theory with rich connotation.Now the clinical workers of massage use the theory—“After Pressing,It Heat Up” into clinical practice,combine the impact of Promoting Qi,Activating Blood,Warming Meridian to Expel Cold and Tonify Deficiency to the treatment of various diseases in the department of traumatology,internal medicine,gynecology and pediatrics.Scientific researchers of massage have explored the theoretical essence of the theory of “After Pressing,It Heat Up” from the aspects of human physiology,pathology and cellular molecular mechanism based on the clinical effect of massage.By analyzing ancient literature of Massage Technique of pressing's operation,the size of the power,the direction of operation,and the way of operating are the primary factors to take into account,the finger temperature is the indispensable condition of action of press,thumb shape and texture are also associated with it.In addition to the principle of mechanics,the nature of human operating method is also needed to be fully understood when designing a stimulator machine of action of press that matches the characteristics of Traditional Chinese Medicine.In our design,we're not only analyzed the laws of kinematics and dynamics of the press,but also fully took the human factors contained in the method into consideration.The kinematics and dynamics parameters of the press method mainly involve the control of force size and direction and the choice of action mode.The characteristics of human operation mainly include the shape and texture of the thumb and the temperature of the finger belly.Objective:1.Explore the influence on heating effect of finger-pressing force and thumb temperature;2.Observe the influence rule of heating effect caused by finger-pressing different force,time and total operating time;3.Observe the influence on heating effect of different finger-pressing parameters,and explore the optimal combination of pressing force,operation time and frequency;4.Observe the influence on heating effect of different finger-pressing operation modes,and explore the optimal operation mode of the finger-pressing on heating effect.Method:1.24 volunteers were divided into three groups,each group separately pressed by thumb direct pressure(pressure + temperature),tightly against the skin by thumb abdomen(temperature),pressed the left-side Xinshu by thumb with insulating sleeve(pressure),observed the variation of local temperature before and after pressing point,gaind the influence on heating effect that caused by the thumb temperature and strength,to verify the scientificity of the raise of ancient theory ”Zhua Ku Shou Du”;2.24 volunteers were divided into three groups,finger-pressing the Xinshu on the left side with different strength(very mild,mild,moderate,severe and severe),different residence time(2 s,4 s,6 s,8 s,10 s),the different total press time(2.5 min,5 min and 7.5 min,10 min,15 min),observed its local temperature changes around the point,to obtain the dose-effect relationship between strength,residence time,total operating time with the heating-effect on pressing;3.Orthogonal test screens the best parameters of heating effect on pressing: refer to the former part of the experiment and literature,several pressing parameters were selected,namely the power(1.5 kg,2.0 kg,2.5 kg),frequency(7.5 points,10.0 points,10.0 times/min),the operating time(2.5 min,5 min and 7.5 min),and then according to the L9(34)orthogonal table of test sequence of three factors three levels orthogonal experiment,the six volunteers as subjects was pressed the Xinshu on the left side with Traditional Chinese Medicine stimulator,the local temperature changes around the point was observed,and concluded the best parameters on heating effect.17 volunteers as subjects were respectively pressed by Traditional Chinese Medicine stimulator with rhythmically press(optimal parameters of part 2.3),continuous press(the optimal strength and total pressurein in section 2.3,remain for 1 min),observe the local temperature changes of the point,thus can conclud the best way of stimulation on “It Heat UP”.Rsult1.The temperature rose by 2.33 ? after thumb abdominal directly pressing,rose by 1.94 ? after thumb abdomen tightly against skin,rose by 1.47 ? after thumb abdominal pressing with insulating sleeve;2.The immediate temperature changes with different strength were 1.88 + 0.64,2.05 + 0.68,2.25 + 0.59,2.35 + 0.61 and 2.32 + 0.69,and the temperature changes after pressing for 15 minutes were-0.11 + 0.11,0.03 + 0.14,0.59 + 0.58,1.38 + 0.70,and 2.09 + 0.98.The immediate temperature changes with different pressing time were 1.94 + 0.37,2.33 + 0.29,2.49 + 0.31,2.51 + 0.39,2.41 + 0.55,after pressing for 15 minutes were 0.53 + 0.49,0.33 + 0.30,0.52 + 0.33,0.55 + 0.38,0.76 + 0.36.The immediate temperature changes after pressing with different remaining time were 2.21 + 0.48,2.37 + 0.46,2.26 + 0.51,2.33 + 0.57,and 2.47 + 0.47.3.Among the three factors of strength,operation time and frequency,the influence of different forces on temperature difference is significantly different(F=32.843,P < 0.05),and the influence of 2.5kg is the most significant.The effect of different operation time on temperature difference was significantly different(F=54.102,P < 0.05),and the effect was most significant at 7.5min.The effect of frequency on temperature difference was not statistically significant(F=2.181,P > 0.05).4.The effect of rhythmic pressure on temperature difference was better than that of continuous press(P < 0.05),but the difference in duration between the two was not statistically significant(P > 0.05).Conclusion:1.Both thumb pressure and thumb temperature have effects on the heating effect,and the heating effect of the combination is the best;2.(1)with the increase of pressure,the temperature rises after pressing immediately,when reach the moderate pressure,the temperature rises by 2.25 ?,close to the stable,but with the increase of pressure,duration of heating effect also grows;(2)as the prolongation of pressing time,the temperature rises after pressing immediately,to 2.49 ? after 7.5 minutes,nearly smooth;(3)The effect of different time on local temperature is not obvious.Objective:1.Observe the effect of pressing Xinshu acupoint on myocardial injury in MIRI model rabbits;2.Observe the effect of pressing Xinshu acupoint on the content of adenosine and HSP70 in myocardial tissue in the infarction area of MIRI model rabbits;3.Observe the effect of pressing Xinshu acupoint on the kinase related to RISK pathway of MIRI model;Method:Through copying the model of ischemia reperfusion injury by recovering blood supply for 3 hours after ligating the left anterior descending artery of the coronary artery for 40 min,we set up the normal group,model group,sham operation group,model + pressing Xinshu group and model + pressing Dazhu group by applying pressing Xinshu acupoint with Traditional Chinese Medicine as intervention and setting contrast of pressing Dashu.Before the experiment,the model group,model + pressing Xinshu group and model + pressing Dazhu group were given modeling treatment,while the sham operation group was only provided with thoracotomy without ligation of coronary arteries.If the model was successful,the two groups,D and E,were respectively given the method of pressing Xinshu point and Dazhu.Electrocardiogram was performed at ligating after and before 10 min and reperfusion 2h to observe ST elevation.Serum CK-MB and c Tn I contents were measured by heart blood sampling before ligation and reperfusion after 3h.After reperfusion for 3h,HE staining and immunohistochemistry of Akt,p-Akt,ERK1/2,p-ERK1/2,PI3 K,p-PI3 K and HSP70 were performed on some of the heart specimens,and adenosine content was performed on the myocardial tissue samples in the infarction area under left anterior descending ramus ligation,as well as on Akt,p-Akt,ERK1/2,p-ERK1/2,PI3 K,p-PI3 K and HSP70 protein Western Blot.Rsult:1.The rabbit ST segment of ECG of model group,model + pressing Xinshu group and model + pressing Dazhu group were all elevated after ligation and decreased after reperfusion,but The difference was not statistically significant;And serum c Tn I increased in all three groups,with the highest increase in the model group.And serum CK-MB increased in all three groups,with the highest increase in the model group,and the difference was statistically significant.2.The contents of adenosine in ischemic myocardial tissues were increased in both model +Xinshu group and model + Dazhu group,and the difference was statistically significant compared with model group.The expression of HSP70 increased in the model group,the model + pressing Xinshu group and the model + pressing Dazhu group,but the difference was not statistically significant.3.The model group,model + pressing Xinshu group and model + pressing Dazhu group can improve the immunohistochemical and WB protein detection of p-PI3K/PI3 K,p-Akt/Akt,p-ERK/ERK ratio,but model + pressing Xinshu group and model + pressing Dazhu group improve more;And model + pressing Xinshu group of immunohistochemical p-Akt/Akt ratio increase more,model + pressing Dazhu group of WB detection of p-ERK/ERK protein ratio improve more.Conclusion:1.Both pressing Xinshu and pressing Dazhu can reduce ischemic reperfusion injury;2.The myocardial protective effect of HSP70 protein can be produced by pressing Xinshu and pressing Dazhu;3.Both pressing Xinshu and pressing Dazhu can stimulate the RISK pathway to exert myocardial protective effect by inducing endogenous triggering substance adenosine.
