Background and Aim Mechanism of bariatric surgery includes caloric restriction and changes in the enterohepatic circulation of bile acids,as bile acids enhance energy expenditure in brown adipose tissue which is associated with weight loss.The aim is to investigate whether bile diversion can reduce body weight,improve liver metabolism,enrich beige adipocytes and enhance brown/beige adipose-driven adaptive thermogenesis in diet-induced obese rats.Methods We performed in vivo studies using our guiding catheter-mediated bile diversion rat model.Bile diversion or sham surgery was performed on diet-induced obese rats.Animals were euthanized 4 weeks after surgery for tissue analyses.Results Significant improvements in metabolic remission and increased plasma bile acid level were observed as bile flow diverted through guiding catheter to ileum.Including sustained improvements in hepatic steatosis and particularly for enhancement of brown/beige adipose-driven adaptive thermogenesis.Conclusions These findings demonstrated that bile diversion led to significant weight loss accompanied by adipose-driven adaptive thermogenesis.These metabolic improvements recommended bile diversion as a new method of treating obesity and metabolic syndrome.
