Objective: To investigate the level of serum G protein-coupled bile acid receptor 1(TGR5)on the population of type 2 diabetes(T2DM)and grasp the trend of TGR5 on the population of T2 DM with or without obesity.In addition,to analyze the correlation between TGR5 with blood glucose,blood lipids,insulin resistance and thyroid function,to explore the potential relationship of TGR5 with glucose metabolism,lipid metabolism and energy metabolism.Methods: Subjects were enrolled from newly diagnosed T2 DM populations who came from health screening at health examination Center of Hebei Province General Hospital from March to May,2017.A total of 105 people(53 males and 52 females)were enrolled in this study,including the control group(CON,37 subjects),overweight group(OW,36 subjects)and obesity group(OB,32 subjects).General information of the subjects was measured,such as height and weight.Blood glucose,blood lipids,serum insulin,total bile acid and thyroid function were measured.Serum TGR5 level was detected by enzyme-linked immunosorbent assay(ELISA)kit.SPSS21.0 software was used for data analysis.Results:1 Comparison of the baseline data among the three groupsCompared with the CON group,BMI,waist hip rate(WHR)and systolic blood pressure(SBP)increased in OW group(P<0.001,P=0.010,P=0.042);BMI,SBP,diastolic blood pressure(DBP),WHR increased significantly in OB group(P < 0.001,P=0.007,P=0.001,P < 0.001).Compared with the OW group,BMI,WHR and DBP increased in OB group(P<0.001,P<0.001,P=0.046).2 Comparison of biochemical index2.1 Comparison of blood glucose related indexCompared with the CON group,fasting insulin(FINS),2 hours postprandial insulin(2h INS)and homeostasis model assessment of insulin resistance(HOMA-IR)increased sharply in OW group(P=0.003,P=0.011,P=0.002)and OB group(P<0.001).Compared with the OW group,Fins,2h INS and HOMA-IR increased rapidly in OB group(P=0.001,P=0.015,P<0.001).2.2 Comparison of blood lipids related indexThe results showed that compared with the CON group,triglyceride(TG)increased dramatically in OW group and OB group(P=0.004,P<0.001),and high-density lipoprotein cholesterol(HDL-C)increased in the OB group(P=0.009).2.3 Comparison of serum total bile acid(TBA)There were no statistical differences in serum TBA among the three groups(P>0.05).2.4 Comparison of thyroid functionThere were no statistical differences in thyroid function among the three groups(P>0.05).3 Analyze the level of Serum TGR5 on the Population of T2DM3.1 The serum level of TGR5 on T2 DM with different body mass indexThe results indicated that the level of serum TGR5 in OB group increased significantly than CON group and OW group(P=0.013,P=0.038).There were no statistical differences in the level of serum TGR5 between CON group and OW group(P>0.05).3.2 The serum level of TGR5 according to the genderThe results showed that there were no significant differences in serum levels of TGR5 between males and females(P>0.05).4 The correlation of serum TGR5 level with general clinical and biochemical dataIn this study,The correlation of serum TGR5 level with blood glucose,blood lipids,TBA and thyroid function was calculated.The results suggested that serum TGR5 level was positively associated with BMI(r=0.298,p=0.002)and 2h INS(r=0.292,p=0.003).5 The influence factors of serum TGR5 levelThe influence factors of serum TGR5 level were analyzed.TGR5 was considered as the independent factor,and multiple linear stepwise regression analysis was used to detect the influence of blood glucose,blood lipids,TBA and thyroid function,the results suggested that BMI was the only important influence factor of serum level of TGR5(P=0.009).Conclusion:1 The level of serum TGR5 in OB group increased significantly than CON group and OW group.2 The serum TGR5 level was positively associated with BMI and 2h INS,and BMI was the only important influence factor of serum level of TGR5.
