detect the expression of mi RNA let-7f in tumor tissues and normal brain tissue samples from glioma patients.The serum mi RNA let-7f expression level of 78 patients with glioma and 51 healthy controls were detected,too.2.The correlation between the expression level of glioma patientsserum mi RNA let-7f and patients’ gender,age,tumor size,site,KPS score before first treatment and WHO pathological grading clinical characteristics was analyzed by single factor and multivariate analysis.3.To analyze the differences ofglioma patients serum mi RNA let-7f expression level in different groups,ROC curve was useed to assess the early diagnostic value of serum mi RNA let-7f.4.All follow-up of gliomapatients lifetime was statistics,the patients’ serum mi RNA let-7f expression and progression free survival(DFS)and overall survival(OS)correlation was analysied,Kaplan-meier method was used to analysis patients serum let-7f value for the forecast of its prognosis.6.Using single variable and multivariate Cox regression analysis to determine whether the serum mi RNA let-7f expression level is an independent factor in the prognosis of gliomapatients,and the evaluation value of serum mi RNA let-7f was discussed.Result1.The expression level of 78 cases of glioma patients serum mi RNA let-7f in the tumor tissue samples was significantly lower than the normal brain tissue(0.35±0.17VS1.12±0.24),there was significant difference(P<0.01).The expression of glioma patients’ serum mi RNA let-7f was significantly lower than serum expression levels of 51 healthy controls(0.39±0.25VS0.89±0.27)(P<0.01).The let-7f expression levels of tumor tissues and their corresponding serum samples in glioma patients were correlated.(r=-0.856,P<0.05).2.Single factor analysis of glioma patients serum mi RNA let-7f expression level may be related to patients’ tumor size,preoperative KPS score,WHO pathology grade and other clinical characteristics(P<0.05).it not related to the patients gender,age and tumor location(P < 0.01),the expression level of glioma patients serum mi RNA let-7f was significantly correlated with tumor size,KPS score and WHO pathology grade according to the result from further logistics analysis(P < 0.05).3.The average expression level of mi RNA let-7f 0.51 for the world is divided into high and low level expression group,WHO grade III-IV glioma patients accounted for66.67%(28/36)of the low level expression group,The I-II glioma patients accounted for 33.33%(14/36)of the low level expression group,There were significant differences between the two groups(X2=8.62,P<0.05).There was significant difference between the expression level of serum mi RNA let-7f in patients with different WHO grades.The higher the WHO grade,the lower the average expression level of the serum mi RNA let-7f(P<0.05).The ROC curve shows the corresponding sensitivity and specificity were respectively 83.78% and 85.71%.That serum mi RNA let-7f level was used to analyze the early diagnostic value of high grade glioma,the corresponding sensitivity and specificity were respectively 83.78% and 85.71%,the early diagnosis and analysis of low grade glioma the corresponding sensitivity and specificity were respectively 94.87% and 85.71%,Lower than all glioma or high grade glioma,as well as the area under the curve(P < 0.05)4.The low expression level of 78 cases of glioma patients serum mi RNA let-7f were significantly shorter than those with high expression group(P<0.05).Patients in WHO classification with low expression level of serum mi RNA let-7f were significantly longer than those with high expression group(P<0.05).6.single variable and multivariate Cox regression analysis were used to determine the expression level of patientsserum mi RNA let-7f,That KPS score and WHO pathology grade were independent factors of PFS and OS in glioma patients(P<0.05).ConclusionThe expression level of glioma patients serum mi RNA let-7f was correlated with the expression level of tumor tissueslet-7f,the clinical features of the patients’ KPS score and the WHO pathology grade were related,it is highly sensitive and specific for people to use serum let-7f in the prognosis of glioma,It is the independent factor of patients PFS and OS,which can be used as a reference index for evaluation of patients survival prognosis. |
