Objective Through in vitro cell function experiment and detect clinical specimens of human breast cancer to study the role of ADAM29 in the development process of breast cancer,in order to provide new ideas and new targets for breast cancer diagnosis and treatment.Materials and Methods In this study,we used MDA-MB-231 cells with differing ADAM29 expression and assessed the influence of ADAM29 and it's gene mutation C92T:P31L,C187T:H63Y,C673A:L225I on MDA-MB-231 cells.We also assessed the expression levels of ADAM29 using immunohistochemical staining assay and Q-PCR in a cohort of human breast cancer specimens.Results 1,Using over-expression and knockdown approach,we created sublines from MDA-MB-231 cells and further demonstrated that ADAM29 expression in MDA-MB-231 cells significantly influenced the aggressiveness of the breast cancer cells,by promoting their cell growth,migration and invasion(P<0.05);2,Mutations in different domain of ADAM29 affect proliferation,migration and invasion of breast cancer in vitro(P<0.05);3,Immunohistochemical staining revealed the increased staining of ADAM29 in breast cancer sections compared with normal tissue(P<0.05);4,Increased transcript expression of ADAM29 was also observed in the breast cancer tissue sections compared to normal tissues(P<0.05),ADAM29 expression was increased in higher grade and higher TNM staging breast cancer,and higher levels correlated with a poorer outcome(P<0.05).Conclusions Cell function in vitro experiments confirmed ADAM29 promotes cell proliferation,migration and invasion in breast cancer.Found ADAM29 mutations that alter its impact on breast cancer cell function.Clinical research is a further indication that ADAM29 plays a promotive role in the development of breast cancer.Provides support for ADAM29 as an important marker to breast cancer diagnosis,treatment and prognosis.
