Grape is an important economic crop in China's agriculture.The aroma trait in grape is a key factor for fruit quality.The genetic mechanism complex and affected by many factors such as variety,environment and cultivation measure.Previous studies mainly focused on the aroma substances and related genes of muscat flavor grapes,relatively few conducted on the composition of aroma substances and related loci of labrusca flavor grape.In order to explore the main aroma substances and candidate genes in labrusca flavor grape,provide theoretical and technical support for analysis of aroma regulation mechanism and variety improvement in the future.In this study applies headspace solid-phase microextraction combined with GC-MS technology quantitatively analyzed volatile component in grape berries of different flavor grape types: ‘Red Globe' ‘Venus seedless' and ‘Muscat hamburg'.We used ‘Red Globe' as female parent,‘Venus seedless' as male parent created a hybrid population and analyzed the inheritable character of main aroma substances.Construction of a highly saturated genetic map and QTLs of major aroma substances were mapped.A number of candidate genes and molecular markers related to main aroma substances of labrusca flavor grape were found.The main results are as follows:1.Construction of a highly saturated,high quality genetic map for vitis by next-generation restriction site-associated DNA sequencing.There were 51,265 markers in the female map ‘Red Globe'(V.vinifera L.)and the total length was 3172.33 c M with an average marker interval of 0.11 c M.The map of the male parent ‘Venus seedless(V.vinifera × V.labrusca)' contained 23,683 markers spanning 3221.4 c M with an average genetic interval of 0.28 c M.The integrated map contained a set of 70,061 markers spanning 3014.46 c M with an average inter-marker distance of 0.05 c M.2.Through principal component analysis(PCA),methyl salicylate,dimethyl phthalate,dibutyl phthalate,damascenone and phenylethyl alcohol flavor compounds were considered as the characteristic flavor compounds in labrusca flavor grape ‘Venus seedless'.The main accumulation periods of different aroma substances are different.3.To analyze the genetic characteristics of the main aroma components of ‘Red Globe' and ‘Venus Seedless' interspecific hybrid progeny for two consecutive years.Methyl salicylate,dimethyl phthalate,dibutyl phthalate,damascenone are characterized by the genetic characteristics of quantitative trait.The intermediates of the esters are lower than the parental mean,the intermediate value of the damascenone is greater than the average of the parents.Phenethyl alcohol showed genetic trait of the quality trait and showed different genetic patterns in two years.4.QTL mapping five main aroma substances of labrusca flavor grape,and 48 QTL loci were located.Analyed the 120 genes in the QTL confidence interval,24 genes may be related to aroma were screened out.Expression analyzed showed that ACP ?,AT-hook7 and COBRA10 may be related to the aroma components of labrusca flavor grape.Predicted and analyzed the basic properties and protein structure of candidate genes shows that ACP? and COBRA10 proteins encoded were hydrophilic proteins with good flexibility located on the cell membrane,AT-hook7 proteins encoded was hydrophilic proteins with good flexibility located in cytoplasm and nuclei.It provides a theoretical basis for the subsequent study on the molecular mechanism of aroma substances in labrusca flavor grape.
