Establish A Knowledge-Driven Migration Learning Method To Compare The Biological Characteristics Of Sperm Function In Multiple Species

With the continuous development of social economy,animal reproduction science has become more and more important in animal husbandry and animal production.Increasing the reproductive rate of animals can continuously expand the coverage of improved varieties,improve animal production efficiency and increase economic benefits.Among them,the study of sperm biology characteristics is the core of animal reproduction,which is closely related to the continuation and genetic evolution of species.With the development of high-throughput sequencing technology and bioinformatics technology,we have obtained relatively detailed genomic information such as gene function,gene expression and mutation detection sites in sperm biology based on a large number of published literatures.However,the prior knowledge of these gene functions and cell biology are mainly concentrated in humans or some model animals,so the distribution of knowledge in different species is unbalanced.When we identified a subject as a certain species,there was no reliable and sufficient data to support the biological study of this species.In order to solve this problem,we constructed sperm biology characteristics knowledge maps of multiple species to achieve the migration of sperm biology characteristics knowledge based on the published sperm biology literatures,comparative genomics data,functional genomics data,and knowledge-driven automated literature mining method.The specific research contents and results are as follows:(1)Using the literature abstract of the keyword "Public Health" as a control group,construct a knowledge map of the biological characteristics of "sperm".Based on the knowledge-driven automated literature mining statistical method,1195 specific genes related to sperm biological characteristics,2162 specific entities,and the correlation matrix between 1195 genes and 2162 entities were obtained as the standard model.At the same time,we obtained the 89 sperm biological characteristics entity categories and the most significant literature abstracts associated with each entity category based on the standardized knowledge classification.(2)Combined with the signaling pathway databases of the IPA(Ingenuity Pathway Analysis)and KEGG(Kyoto Encyclopedia of Genes and Genomes),the signaling pathways significantly related to the sperm biological characteristics entity categories(i.e.sperm biological characteristics categories-signaling pathways)and the genes enriched by the signaling pathway are obtained.Further analysis,we can obtain a significantly activated signaling pathway and a significantly inhibited signaling pathway corresponding to each sperm biology characteristics category,and the sperm biology characteristics category corresponding to each signaling pathway being significantly activated and significantly inhibited for each of the 187 signaling pathways.(3)To construct the knowledge map of sperm biological characteristics of Mongolian five animals "Capra hircus,Ovis aries,Camelus bactrianus,Equus caballus,Bos taurus " based on species genomics data and homologous genomics data.The migration of sperm biology knowledge between species is carried out in combination with the human sperm biology standard knowledge database and the signaling pathway function database.We found that 10 sperm biology characteristics category_signaling pathways were significantly conserved among 5 species(Z-score>5),as well as specific signaling pathways that were significantly conserved for different species(z-score>3).At the same time,in the gene network enriched by the relatively conserved 110 sperm biology characteristics category signaling pathways(z-score>3),the RAF1,GRB2,and EGF gene were found to be pivotal genes in the network.(4)The mobility of sperm biology characteristics was assessed between two pairs of allied species,"Equus caballus and Equus asinus,Goat and Sheep",and each pair of allied species were significantly conserved and sperm biology characteristics category_signaling pathway that can migrate with each other.And the biology characteristics category_signaling pathway between allied species in which a single species is significantly and specifically conserved and non-migratory(Z-score>5).(5)The mobility of sperm biological characteristics was evaluated among 4 Camelidae species,"Camelus dromedarius,Camelus bactrianus,Vicugna pacos,Camelus ferus".A list of 13 sperm biology characteristics category_signaling pathway that is significantly conserved and migrates among 4 species,as well as single species specifically conserved and non-migratory sperm biology characteristics category_signaling pathway(Z-score>3).In addition,we obtained the sperm biological characteristic category_signaling pathway was not significantly activated only in a certain species.As the "Toll-like Receptor Signaling" signaling pathway was not significantly activated in the biological characteristics associated with the DNA structure of the spermatozoa of Vicugna pacos,but was significantly activated in Camelus dromedarius,Camelus bactrianus,and Camelus ferus.Therefore,we use the abstract of sperm biology literature as a knowledge background to construct a knowledge-driven automated literature statistical mining method to draw a standard knowledge map of sperm biology.At the same time,the migration learning method was used to construct a multi-species sperm comparison functional biology group,and then the mobility of the sperm biological characteristics between species was evaluated,which provided the knowledge and theory basis for understanding the sperm biological characteristics of different species.
