Study On Quality Of Lonicerae Japonicae Flos New Variety "Huajin 6"

Objective: Under the guidance of the traditional theory of traditional Chinese medicine,combined with modern scientific and technological means,the new honeysuckle variety“Huajin 6” with an extended flower bud stage was selected,in order to provide scientific basis for its next promotion,this paper systematically evaluated the quality of “Huajin 6” based on traditional honeysuckle as a control.Methods: Using systematic breeding combined with radiation induction,a new variety of honeysuckle “Huajin6” with an extended flower bud stage was selected.This paper observed the original plant morphology and growth and development habits,medicinal properties and microscopic characteristics.HPLC and GC-MS analysis was used to compare the difference of active ingredients between "Huajin 6" honeysuckle and other varieties,and HPLC fingerprint of "Huajin 6" was established.In order to select the appropriate harvesting and processing methods,we collected “Huajin 6” samples with different developmental stages and different drying methods,the morphological characteristics,chemical composition,fingerprints,volatile oils,etc.were compared and analyzed.The ITS2 gene fragments of “Huajin 6” honeysuckle and other varieties were extracted,combined with the ITS2 gene fragment of Lonicera confusa from Gen Bank,the basal origin and phylogeny of this species were analyzed at the molecular level.Studied the acute toxicity of "Huajin 6" honeysuckle aqueous extract on mice;compared the differences of antiviral and antioxidative abilities from different polar parts of the variety in vitro,and screen out the best active site.Results:(1)We successfully selected "Huajin 6" honeysuckle,its branches are prosperous,strong in erection,bud hypertrophy and growth concentrated,the white bud stage lasts for 15 to 20 days.The yield is 1.43 times that of traditional variety,and the contents of chlorogenic acid and cynaroside are 2.03 and 2.08 times that of the Chinese Pharmacopoeia respectively.(2)Compared with traditional varieties,the leaf shapeof "Huajin 6" are oval,with dense white hairs on the leaves.The pistil and stamens are lower than the corolla.Flowering period late,the white period lasts longer in the process of flowering.The flower buds are directly opened to the golden flower period,and lack of the silver flower period.(3)Herbs color yellowish white;buds larger,vacuolated,slightly cracking upper part;some flower buds lack of calyx,has dense glandular hairs and non-glandular hairs,and glandular hair glands with large head and short stem.(4)The content of total flavonoids and total iridoid glycosides in this variety was significantly higher than other varieties,especially in the content of cynaroside and loganic acid,which were 2.26 and 3.92 times of “Huajin 2” respectively;There was no significant difference in the composition of volatile oil components and other varieties,but the contents of the volatile oil was significant difference.The HPLC fingerprint of “Huajin 6” was established,which can better evaluate the quality of “Huajin 6”medicinal materials and quickly identify the variety and other varieties of honeysuckle and lonicerae flos.(5)The quality of "Huajin 6" was stable during the white period and is suitable for picking.There are significant differences in the quality of herbs in different drying methods,and the quality of drying samples is the best.(6)ITS2 gene fragment can distinguish “Huajin 6” from other varieties of honeysuckle,and the phylogenetic relationship between this species and Damaohua is closer.(7)"Huajin 6" was safe to use within a safe dose.Compared with the Damaohua,the extract of “Huajin 6” has stronger effects of anti-HSV-1,especially the water-extracted and alcohol-sinking sites.The 50% ethanol and 70% ethanol extract sites showed strong DPPH antioxidant activity.Conclusion: Compared with traditional varieties of honeysuckle,"Huajin 6" not only has significant advantages in appearance and intrinsic active ingredients,but also has a stable quality throughout the big white periods,which can significantly extend the picking time and reduce the cost of harvesting.Its clinical use is safe and its pharmacological activity is remarkable.Comprehensive analysis shows that the variety is suitable for clinical application and large-scale promotion of planting.The successful cultivation of "Huajin 6" honeysuckle has brought new opportunities to the honeysuckle industry.By systematically studying the quality of its medicinal materials,it provides basic information for the further promotion and the development of new products.
