Innate lymphoid cells(ILCs)are the most recently identified family of innate immune system that believed to modulate immune functions prior to the generation of adaptive immune response.While,subsets of ILCs reside in the mucosa and regulate immune responses to external pathogens,their role and mechanism in protecting against intracellular bacteria infection is incompletely understood.In this report,by establishing S.Typhimurium and L.monocytogenes infection model,we found that the cell number,associated with the Runx3 expression,of group 1 ILCs and NCR+ ILC3s were increased.Besides,Runx3fl/fl PLZF-cre+/-mice were much more sensitive to the infections of intracellular bacterial S.Typhimurium and L.monocytoge due to the cell intrinsic reasons of abnormal Group 1 ILCs and NCR+ILC3s.In addition,we found that Runx3 could directly bind to ill2r?2 promoter and intron 8 to accelerate the expression of il12r?2 and modulate IFN? secretion triggered by 'IL12/STAT4' axis.Thus,we demonstrate that Runx3 is participated in group 1 ILC and NCR+ILC3 involved immune protection to intracellular bacterial infections in gut and liver.
