Objective: To construct indicator system of talented identification for Chinese female long-distance runners at advanced stage, which was combined with gene polymorphisms, by testing the level of morphology,physiology, psychology, sports quality and strategy.Method: (1)By measuring 101 female long-distance runners in terms of primary indicators, and expert questionnaire, factor analysis and regression analysis were used to select suitable indicators and construct comprehensive evaluation standard of talented identification. (2)Using polygenic calculation method to analysis polygenic characteristic from gene polymorphisms associated long-distance running performance, using logistic regression to construct genetic predicted model for elite performance, using AUC to test diagnostic capability of predicted models based on different genetic effect, using comparison of ROC to determine the best model. (3)Using MALDI-TOF technique to genotype 11 gene polymorphisms identified in study 2. Then there were two ways to be choosed. One was to select athletic performance phenotype as dependent variable, indicators identified in study 1, combined or not combined gene polymorphism as independent variables, using logistic regression to construct qualitative evaluation system. The other way was to select score from study 1, total genotype score and its relative weights from study 2 to calculate new score, which was combined with gene polymorphisms to construct quantitative evaluation system. Using comparison of ROC to determine the difference of diagnostic capability of Indicator System combined or not combined gene variants.Result: Morphology indicators selected in system were the ratio of crista breadth and hip width, the ratio of lower limb length A and height, the ratio of lower limb length A and H. Physiology indicators were velocity at anaerobic threshold, ventilation, heart rate and maximal oxygen uptake achieved in an incremental loading exercise test, and hemoglobin.Psychology indicators included scorings of perseverance, independence,consciousness and decisiveness. Sports quality indicators were 1500 meters performance and functional movement screening scorings. Rhythm,tactics and anxiety were selected in term of strategy indicators. Total genotype scores and the accordance rates of advantageous genotype in elite athletes were significantly higher than those in controls (P<0.01,P<0. 01), which suggested athletes presented better polygenic profiles.A genetic predicted model based on advantageous effect with a Nagelkerke' s pseudo R2 of 0.291 gave best diagnostic capability with a AUC of 0.78 to differentiate athletes from controls(P<0. 01). AUC of quantitative way increased from 0. 73(P>0. 05) to 0. 82(P<0. 01) in national team when combined total genotype score, and AUC of qualitative way increased from 0. 84(P<0. 01) to 1.00(P<0.01) when combined genotypes. It was concluded that new indicator system combined gene variants could improve diagnostic capability of talented identification.Conclusion: Traditional indicator system of talented identification for Chinese female long-distance runners and genetic predicted model for Chinese female long-distance runners were identified. Both model presented suitable clinical validity to diagnosis whether one had potential for elite performance or not. New indicator system combined gene variants could improve diagnostic capability of talented identification.
