Part ? The concentration and clinical significance of YKL-40 in serum of prostate cancer patientsObjective:To understood the expression of serum YKL-40 in prostate cancer patients,and analyze the relationship with the occurrence,development,metastasis and prognosis of prostate cancer,aimed to explore the YKL-40 as an index for early screening the prostate cancer,and even as a specific serum laboratory indexes to monitoring and prognosis on prostate cancer.Methods:Collected blood samples from prostate cancer patients,when they before and after treatment.At the same time,choose the prostate hyperplasia patients and healthy controls blood samples men as a comparative object.Used enzyme-linked Immunosorbent(Enzyme.1 inked Immunosorbent Assay,ELISA)double antibody clamp method to detect the concentration of serum YKL-40.According to the clinical indicators such as rectal examination,prostate tissue biopsy,serum prostate-specific antigen(PSA),and postoperative pathological examination,the correlation of YKL-40 was analyzed.Results:1.The concentration of YKL-40 in the serum before treatment of prostate cancer patients was significantly higher than that of normal control group and prostate hyperplasia group,and the content of YKL-40 in serum was significantly lower after treatment than before treatment.2.Compared with PSA,the positive predictive value of serum YKL-40 has no statistically significant difference between,when PSA>10 ng/ml and YKL-40>120.89ng/ml as joint diagnosis reference,positive predictive rate trend for improvement of the diagnosis of prostate cancer.3.When PSA >40ng/ml and YKL-40 >143.69 ng/ml as joint diagnosis reference,used to diagnose prostate cancer with bone metastases of positive predictive rate significantly increased.Conclusion:1.The content of YKL-40 in the serum of patients with prostate cancer after radical prostatectomy and endocrine therapy decreased significantly.It is suggested that detection of YKL-40 has certain clinical significance for the diagnosis and prognosis of prostate cancer.2.PSA as prostate specific antibodies,90% of patients with prostate cancer before treatment the serum PSA abnormal(> 4 ng/ml),and when the PSA and YKL-40 joint diagnosis as a reference,the diagnosis of prostate cancer and to determine the positive predictive rate of bone metastases trend will increase significantly.Part ? Expression and clinical significance of YKL-40 in prostate cancer tissueObjective:Studied in this research by detecting different YKL-40 expression in prostate tissue and analyze clinical data of patients with prostate cancer and tissue in the correlation of the expression of YKL-40,to explore YKL-40 in the progression of prostate cancer and the role of tumor invasive significance,so as to clarify the occurrence of prostate cancer invasion mechanism provides a new train of thought,further evaluation of patients with prostate cancer clinical progress,and provides some references for the treatment effect of the prognosis.Methods:This study collected tissue specimens of surgical resection in the urology department of the first hospital of Shanxi Medical University,including normal prostate tissue,prostate hyperplasia,and prostate cancer tissue.The content of YKL-40 in prostate tissue was determined by immunohistochemistry.Spearman rank correlation was used to analyze the correlation between YKL-40 and Gleason score,clinical stage and serum PSA.Results:1.Optical microscope found that YKL-40 mainly in the diffuse expression of prostate epithelial tissue,cell membrane,nucleus and prostate in the connective tissue and smooth muscle did not see YKL-40 expression.The expression of YKL-40 was significantly increased compared with normal prostate group and prostate hyperplasia group,indicated that the expression of YKL-40 was significantly correlated with malignant hyperplasia of the prostate.2.There was no difference in YKL-40 expression between the age of 75 and 75 above,indicating that the expression of YKL-40 was not closely related to age.In different clinical stages of prostate cancer,? and ? cases YKL-40 positive expression rate of 100%,significantly higher than the ? and ?;YKL-40 positive expression rate was 100% in the low-differentiation group of prostate cancer,significantly higher than that in the middle and high differentiation group.The positive expression rate of YKL-40 in the lymphatic metastasis group of prostate cancer was100%,higher than that in patients without metastasis.The positive expression rate of YKL-40 was 100% in the bone metastasis group of prostate cancer,significantly higher than that in patients with prostate cancer.Conclusion:1.The expression of YKL-40 in prostate cancer tissue is higher than that in normal prostate tissue and hyperplasia.2.The expression of YKL-40 was positively correlated with the pathological grading of prostate cancer,Gleason score and clinical staging.3.The expression of YKL-40 was positively correlated with the invasion of prostate cancer.4.The expression of YKL-40 was not significantly correlated with the age of prostate cancer patients.Part ? YKL-40 promotes the migration and invasion of prostate cancer cells by regulating epithelial mesenchymal transformationObjective:In this study,we compared the expression levels of YKL-40 in prostate cancer tissues and normal prostate tissues.The expression level of YKL-40 in various prostate cancer cells and its correlation with metastasis and invasion ability were analyzed.By up-regulating and down-regulating the expression level of YKL-40 in prostate cancer cells,the regulation of EMT was observed,and the mechanism of YKL-40 in the metastasis and invasion of prostate cancer was elucidated.Methods:This study collected tissue specimens from the surgery of urological prostate cancer in the first hospital of Shanxi Medical University,including prostate cancer tissue and adjacent tissue.The expression of YKL-40 in tissue of immunohistochemistry was determined by Western blot and paraffin section.In addition,the cell lines of YKL-40 overexpression and down-regulated expression in the DU145 cell line were constructed to detect the migration rate and invasion ability.Results:1.Immunohistochemistry and Western blot showed YKL-40 expression level in cancerous tissue is significantly higher than tissue adjacent to carcinoma.2.YKL-40 high expression of DU145 cell migration and invasion can be significantly higher than YKL-40 lower expression of PC3 cells,DU145 cells by lentivirus YKL-40 expression,cell migration and invasion ability;YKL-40 and raised PC3 cells after the expression of cell migration and invasion ability enhancement.Prove YKL-40 expression level is closely related to the prostate cancer cell migration and invasion ability.3.In YKL-40 high expression levels of cells,the E-cadherin expression level was significantly lower and interstitial cell markers N-cadherin and Vimentin expression levels drop.EMT related transcription factor q,Slug and Twist in YKL-40 high expression levels of expression level is higher than in the cells YKL-40 low expression level of cells.These results clearly show that YKL-40 expression level and EMT related gene expression level are related.Conclusion:1.YKL-40 may expression in benign prostate hyperplasia and prostate cancer tissue.The expression level of YKL-40 in prostate tissue is higher than those in BPH and healthy males.2.The expression level of YKL-40 in human prostate cancer DU145 and PC3 cell lines was related to cell migration and invasion ability.3.YKL-40 affects cell migration and invasion by modulating the expression levels of the EMT genes in the DU145 and PC3 cell lines of human prostate cancer.
